Objective: Dendrobium officinale is one of subspecies in genus of dendrobium,which own special potency as one kind of medicine.Since the development of Dendrobium officinale is rapidly,it is urgent to conduct the research of its active ingredient and its relevant mechanisms.The preparation of a new dendrobium officinale reagent with high dissolution could not only make the best use of dendrobium officinale,but also greatly improve the effectiveness as medicine used for organisms.Based on the result of its corresponding physics and chemical characters,this article mainly discussed the technology and relevant criterion about the preparation of dendrobium officinale oral solution.Furthermore,the pharmacodynamics of the oral liquid on hepatic adipose infiltration were also studied.Method: First of all,according to its physics and chemical characters,the preparation of dendrobium officinale included discoloration using 95% ethanol,extraction of tissue homogenate,centrifugation in 10000 r,filtration,subpackage of semi-finished products into 10 ml per tube,and finally obtaining the product after sterilizing in 121 degrees centigrade.The polysaccharide of dendrobium officinale was identified by thin-layer chromatography(TLC).The content of mannose and polysaccharide were measured by high performance liquid chromatography(HPLC),and the relevant quality criterions of were also developed.In addition,the preparation of animal liver injury model was made by feeding by gavage of C57BL/6J mice with Lieber-DeCarli liquid alcohol for 15 days.There are six groups in this model,including Compound Mehionine,Choline Bitartrate Tablets positive control group,dendrobium officinale oral liquid high,medium and low dose groups by gavage,respectively.The blank group and model groups were fed with same solvent(injection water).The mouses were feed for 15 days.Weighing the body weight of mice after the last dose of 2 hours,collecting blood from eyeballs,stationary for 30 minutes at room temperature,3500 r centrifugation for 10 minutes,and taking the supernatant fluid.Pick up the livers of mice,weigh the wet weight,cutting out part of liver tissue and weighing it,9 times diluted homogenate,3500 r centrifugation for 10 minutes and taking supernatant fluid.Besides,taking part of liver tissue for histopathology sections.Detecting the quantity levels of ALT,AST,ALP,TG,TC,HDL-C,LDL-C in serum and TG,TC,HDL-C and LDL-C levels in the liver tissue.The pathological changes(inflammation and steatosis degree)were also observed.6month old HBV(Hepatitis B virus)transgenic mice was utilized(preliminary study found that 5month old HBV transgenic mice could form the lipid droplets accumulation.).The mouse were divided into six groups,silybin meglumine capsule positive control group,dendrobium officinale oral liquid high,medium and low dose groups by gavage,the blank group and 2C model groupsfed with same solvent(injection water).The mouse was conducted by gavage once a day for 60 days.After the last dose,the mouse was weighed.Collecting blood from eyeballs and placing at room temperature for 30 minutes,3500r centrifuged for 10 min and taking the supernatant fluid.Picking up the liver and spleen from the mice,weighing the wet weight,cutting out part of liver tissue and weighing it,9 times diluted homogenate,3500 r centrifugation for 10 minutes and taking the supernatant fluid.Besides,making liver histopathology sections(HE staining and Sirius red staining)and spleen histopathology sections.Detecting the relative expression of AST,ALT,TG,TC,HDL-C,LDL-C,TNF-alpha and IL-6 in liver tissue.The expression of Bax and Bcl-2 were observed by immunohistochemistry.The protective effect and mechanism of dendrobium officinale oral liquid on HBV transgenic mice were studied.Results: The content of dendrobium officinale was strongly characterized in the oral liquid by TLC identification.The polysaccharides and mannose in dendrobium officinale oral liquid have good linear correlation in the range of 0-0.11mg/ml and 0-30mg/ml..Alcoholic liver injury model in mice was successfully established,and dendrobium officinale oral liquid in large and medium and small dose groups could significantly reduce the AST,ALT,TG,TC,gamma-GT,ALP and LDL-C levels,while the content of HDL-C increased significantly.Pathological section represented that the oral liquid can significantly reduce the fat lesions,has certain protective effect to liver.HBV transgenic mouse model of primary fatty liver was successfully established.The oral liquid in large,medium and small dose groups can significantly reduce the quantity levels of AST ALT,TG,TC,TNF-alpha and IL-6 in the liver tissue homogenate,besides,the LDL-C quantity level decreased significantly in the liver tissue homogenate,while the HDL-C quantity level increased significantly,the Bax expression level decreased.These proved that the oral liquid could reduce the probability of fatty liver lesion and the risk of liverfibrosis.Conclusion: The oral liquid of Dendrobium officinale has a certain protective effect on the liver.The mechanism of its action may be related to the inflammatory factors in the body.
