Association Between Connexin-40 Gene-44G/A、+71A/Gpolymorphisms And Atrial Fibrillation In Uyghurand Han Population In Xinjiang

Objective:To elucidate the association between the polymorphisms of connexin-40(Cx40) and atrial fibrillation(AF) in Uyghur and Han population of Xinjiang region.Method: 275 Uyghur and 305 age- and sex-matched Han subjects were enrolled.5milliliter blood was collected for DNA genotyping,and Cx40 gene polymorphisms(-44G/A and +71A/G) were determined using polymerase chain reaction(PCR). Mutation screening was performed by direct sequencing and calculating the distribution frequency of susceptibility genes to determine the relationship between gene polymorphisms and the atrial fibrillation among Uyghur and Han population. Results:We found that the two single-nucleotide polymorphisms(SNPs) were both significantly associated with AF in both Uyghur and Han populations.In pairwise linkage disequilibrium analysis,and the two SNPs were in perfect linkage disequilibrium in both AF group and control group among Uyghur and Han population.In both Uyghur and Han population,the-44 AA genotype and frequency of A allele in the AF group were significantly higher than those in the control group.In addition, genotype analysis was also performed using several genetic models,and it was found that for both Uyhgur and Han population,the dominant model(GG vs GA+AA) of Cx40-44G/A showed a significant difference between the cases with AF and the controls.Logistic regression analysis showed that Cx40 genetic polymorphisms increased the risk of AF in Uyghur and Han residents of the Xinjiang region.Conclusion:Both-44G/A and +71A/G of the Cx40 gene are associated with AF in the Uyghur and Han population in the northernw part of China.
