Objective To investigate the potential association of angiotensin Iconverting enzyme (ACE) gene I/D polymorphism and angiotensinconverting enzyme2(ACE2) gene A9570G polymorphism with atrialfibrillation. Methods A total of305patients were recruited and dividedinto two groups: the atrial fibrillation group (148cases) and control group(157cases). The control group were matched to cases with the age, gender,presence of left ventricular dysfunction, presence of coronary heart disease,presence of diabetes, and presence of primary hypertension. The ACE geneI/D polymorphism and ACE2gene A9570G polymorphism were genotypedwith polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) and gene sequencing approach. Results The genotypedistribution and allele frequencies of ACE gene I/D was not differentbetween the atrial fibrillation groups and control (P=0.841; OR=0.948,95%CI0.680-1.322, P=0.755, respectively). And there was no significantdifference between left atrial dimension or right atrial dimension of different genotypes of ACE gene I/D polymorphism (P=0.887, P=0.664,respectively). In male subgroup, there was no statistical difference betweenthe atrial fibrillation groups and control in ACE2gene A9570Gpolymorphism (OR=1.631,95%CI0.880-3.023, P=0.119). However, in themale with atrial fibrillation subgroup, left atrial dimension and right atrialdimension of subjects with G genotype were larger than that of subjectswith A genotype (37.0±4.4mm vs40.1±6.4mm, P=0.028;36.5±4.4mm vs40.1±5.7mm, P=0.010, respectively). In female subgroup, The genotypedistribution and allele frequencies of ACE2gene A9570G polymorphismwere not different between the atrial fibrillation groups and control (P=0.286; OR=1.415,95%CI0.885-2.264, P=0.146, respectively). In thefemale with atrial fibrillation subgroup, no significant difference was foundbetween left atrial dimension or right atrial dimension of differentgenotypes of ACE2gene A9570G polymorphism (P=0.924, P=0.432,respectively). Conclusion The genotype distribution and allelefrequencies of ACE gene I/D and ACE2gene A9570G polymorphism werenot significant relationship with atrial fibrillation. However, in the malewith atrial fibrillation subgroup, the G genotype in ACE2gene A9570Gpolymorphism may be a risk factor for prediction of atrial enlargement.
