Objective: Establishment of human-flora-associated( HFA) animals related with Kazakh obesity children in Xinjiang based on germ-free(GF) mice, to study the engraftment of intestinal flora in GF mice, to provided a new way for the further study of the physiopathologic mechanism of Kazakh obesity children. Methods: Cultivated fresh dejecta from Kazakh obesity children who were 7-13 years old in Xinjiang, 10 male and 10 female. 30 GF C57BL/6J mice, divided into EG(20) and CG(10), both half male and half female. The EG mice began to inoculating with stool suspension from Kazakh obesity children when they were 3 weeks,following the principle of gender agreement, once every two days, given three times totally,each time 0.1ml, to build the HFA mice model; The CG mice continue regular to raised in a sterile condition, without any handle. On the 1 month after inoculation,fresh feces samples were collected. PCR-DGGE technology was used to test the stool samples of EG, CG and Kazakh obesity children, and analyzed the diversity of microflora in the DGGE profiles, choose the advantages and clear stripe tag in DGGE profiles to recovery/cloned and sequenced, with the purpose of understand the engraftment of intestinal flora in EG mice. Results:(1) In the early stage of inoculating fecal bacteria from Kazakh obesity children, three female mice of EG death, the rest of the mice lived well.(2) EG mice were normal size, the cecum were reduced to the normal after inoculation,few food residue stranded.The cecum of CG mice were significantly expand,saw appendix when abnormal anatomy, full of food residue.(3)Stool negative bacteria detection in CG mice; each samples of Kazakh obesity children and EG mice in PCR-DGGE fingerprint form complex and dense bacterial bands.(4)EG mice average abundance of flora was 0.76 times of Kazakh obesity children, the difference was statistically significant(P= 0.001); theflora diversity index is lower than the Kazakh obesity children, the flora evenness is slightly higher than that of Kazakh obesity children, but there were no statistically significant difference(P=0.278,P=0.278);(5)The advantage bacterium of intestinal flora in Kazakh obesity children, were all measured in EG mice, there were four kinds of common species sequence at least:Bacteroides, Firmicutes, Bif-idobacterium,Escherichia. Conclusion : The intestinal flora of Kazakh obesity children were colonized in EG mice as much as 76 percent, and the main advantage bacterium group were all get well colonized, EG mice simulated the intestinal flora environment of Kazakh obesity children succeeded,building HFA animal models of Kazakh obesity children.
