Background:Cardiovascular disease has become a global public health problem. The prevention and treatment of cardiovascular disease are also emerging in endlessly, the development and application of new technology or the instrument avail the patients with cardiovascular diseases. Positioning dosing instrument based on the iontophoresis theory, pulsed electric field generated by direct-current(DC),assists the ion drug to act on the lesions directionally. The validity of the instrument has confirmed by the vitro experiments. Drug efficacy and safety in irradiated animals become a focus in the study of this article. Objective:The aim of the research was by detecting heart and adjacent organs verapamil concentration change of the aged rats which irradiated by dosing instrument, to explore the function of the irradiation of the positioning dosing instrument with the ionic drugs targeting and focusing on the heart or adjacent organs(lung)efficiently, then providing the new way for prevention and treatment of the targeted drugs used to treat cardiovascular diseases. Method:We choose 32 F-344 BN rats which were 36 months old, divided into four groups randomly, eight rats in each group. According to the different voltage, duty ratio, frequency of the instrument divided into the irradiation group(470V VII 31 1000HZ), the irradiation group(220V 3:1 1HZ), the irradiation group(220V 5:1 4000HZ)and control group without irradiation. The rats were narcotized mildly. Rats after weighing,did limb lead electrocardiogram(ecg) and recorded for 30 minutes,then injected verapamil by the sublingual vein. The two electrodes were placed in chest and back of the rats,irradiation groups were given different illumination conditions, did nothing with the control group. After an hour did limb lead electrocardiogram(ECG) and recorded for 30 minutes. Then draw blood from abdominal aorta to test CK, LDH, AST, K+, and Na+,detecting positioning dosing instrument’s effects on myocardium and ion of the rats. Then put to death in rats, and took its hearts and lung, tested verapamilp concentration of theintermediate organization by the LC method, in order to make clear the effectiveness of positioning dosing instrument. Result: 1. Compared with control group, the ventricular premature beat( VPB) of the aging rats which arirradiated decreasese significantly(P<0.05), and differences in the irradiated group are not statistically significant(P>0.05). 2. The verapamil concentration from myocardium and lung tissue of the irradiated rats are significantly higher than the control group, and differences are statistically significant(P<0.05); In the irradiation group,the verapamil concentration from lung tissue is significantly higher than myocardium, and differences are statistically significant(P<0.05), but in control group, their differences are not significant(P>0.05). 3. In the irradiation group, when the condition is 220 V 5:1 4000 HZ the verapamil concentration from myocardium is significantly higher than the other two groups(P<0.05). In the irradiation group, the verapamil concentration from lung tissue which conditions are 220 V 3:1 1HZ and220V5:1 4000 HZ is not markedly different(P>0.05), but they are significantly higher than the other groups(P<0.05). 4. Myocardial enzyme and blood ion: compared with control group, CK, LDH, AST, K+ and Na+ concentration from irradiation rats’ don’t change significantly, the differences not statistically significant(P>0.05). Conclusion: 1. The Positioning Dosing Instrument can be applied in targeting therapy. 2. After irradiating for a short time(one hour)by DC pulsed electric field,myocardial enzyme, K+ and Na+ concentration of the rats don’t change significantly. 3. The nearer the distance of the irradiated parts from electrodes of positioning dosing instrument is, the higher the drug concentration of organization maybe. 4. In terms of positioning dosing instrument’s parameter Setting, when voltage, duty ratiom and frequency are 220 V 3:1 1HZ,and it is the ideal irradiation condition in this experiment.
