Objective:1. Through the establishment of cold coagulation and blood stasis type of EMS rats, observe the change of shape of the endometriosis ectopic lesions, and changes of Hemorheology and coagulation function,to judge the best time point of the drug intervention on EMS rats.2. Give the EMS rats therapeutic intervention,to observe the effects of the TONG YU SHU particles with the changes on rat ectopic pathological morphology and its effect on OPN expression,o explore the effect of TONG YU SHU particles on the type of cold coagulation and blood stasis EMs, and provide a basis for clinical treatment.3. Give the EMS rats preventive intervention, observe the influence of OPN expression and EMS rat endometriotic lesions of pathological changes,with using prevention dose of TONG YU SHU particles,and provide a basis for preventive medicine.Methods:1. Build up congealing cold and blood stasis Endometriosis Rat Model with the autologous endometrial transplant method,and then soak the rats with mixture of ice and water,and after the rats were modeling 3 days,5 days,4 weeks,6 weeks and 8 weeks,take four rats randomly from each group,observe the its characterization of congealing cold and blood stasis,and at the same time,observe the morphological and pathological changes of ectopic tissus.2. Give the EMS rats therapeutic intervention,to observe the pathological changes of ectopic tissus in each group(normal control group,model 4 weeks group,model 8 weeks group,the TONG YU SHU low-dose group,TONG YU SHU middle-dose group,TONG YU SHU high-dose group,western medicine group) by HE staining,meanwhile observe the effect on OPN expression.3. Give the EMS rats preventive intervention,HE staining was used to observe the pathological changes of each experimental group, the expression of OPN was detected by immunohistochemistry.Results:1.3 days and 5 days groups just showed inflammatory reaction,but the 4 weeks,6 weeks and 8weeks groups showed the mature endometrium tissue.The model group showed the cold-evil appearance,and showed abnormal hemorheology and coagulation function.There were no statistical significance between the blood parameters of each model group.2.After the medicine was poured into the rats’stomach,In model rats of each geoup, the volume of ectopic tissues were reduced, and the epithelial cells atrophy, the arrangement was not continuous, the stromal cells were decreased, and the number of glands decreased.Among them, the TONG YU SHU high-dose group and the western medicine group were both the most obvious groups.The changes of cold-evil appearance in rats were more obvious in each TONG YU SHU groups,but there were not only no change in western medicine group,but also appeared the adverse effects of weight gain.The blood parameters of rats in each model group were obviously improved by the TONG YU SHU particles group.There were no statistical significance between the western medicine group and the model group.Compared with the blank control group, the expression of OPN in the model 4,8 week group and each drug treatment group was significantly higher, and the difference was statistically significant (P<0.05).Compared with the model group, the expression of OPN in each drug treatment group was significantly lower than that of the model group, and the expression of OPN was significantly higher in the 8 week group compared with the model 4 week group, the difference was statistically significant (P<0.05);Compared with the 8 week group,the expression of OPN decreased significantly in the TONG YU SHU group and the western medicine group, and the difference was statistically significant (P<0.05);compared with western medicine,the expression of OPN decreased significantly in the TONG YU SHU high-dose group,and the difference was statistically significant (P<0.05).3.In the preventive medicine experiment,there was no obvious syndrome of cold-evil in prevention group,and the volume of ectopic tissue was reduced, compared with the TONG YU SHU high-dose group,the interstitial cells were less in the prevention group,the deformation and reduction of the glands was obvious,the degree of fibrosis was higher, and there was a degenerative change.Compared with the blank control group, the expression of OPN was significantly increased in the 4 week group and the TONG YU SHU high-dose group, and the difference was statistically significant (P<0.05);compared with the 4 week model group,the expression of OPN was significantly lower in the TONG YU SHU high-dose group and the prevention group,and the difference was statistically significant (P<0.05);Compared with the TONG YU SHU high-dose group, the expression of OPN in the prevention group was significantly lower,and the difference was statistically significant (P<0.05).Conclusion:1. In this experiment,we use utologous endometrium transplantation method and ice water immersion successfully establish the type of cold coagulation and blood stasis endometriosis model,dynamically observe the change of shape, pathology, blood rheology and blood coagulation function of ectopic foci in Ems,the start time of prophylactic administration was determined on the 5 day after modeling,the time point of therapeutic drug administration was 4 weeks after modeling.2. Compared with the normal endometrium,the expression of OPN in blood stasis type of endometriosis in ectopic endometrium of rats increased significantly,its abnormal expression may plays an important role in the occurrence and development of endometriosis.3. The TONG YU SHU particles can effectively inhibit endometriosis disease,and make the ectopic tissue atrophy and degeneration,and in the improvement of Endometriosis Rat Blood Stasis characterization, blood rheology and coagulation function, the effect is obvious,and ite also have no side-effect like western medicine.It also can restrain the expression of OPN in ectopic tissue, the progress of endometriosis disease can be suppressed by the inhibition of expression in OPN.The high-dose TONG YU SHU is the best dose for treatment of cold coagulation and blood stasis type of endometriosis.4. TONG YU SHU particles of preventive medication can lead to ectopic lesion tissue degeneration and loss its normal form,and it can also inhibited the expression of OPN,and its therapeutic effect was better than therapeutic drugs,theTONG YU SHU particles have early prevention and treatment of endometriosis.
