Objective:Vespae Nidus is the nest of Polistes olivaeeous(De Geer), Polistos joponicus Saussure or Para Polybiavaria Fabrieius which belongs to inseets of vespidae. Nidus Vespae is widely distributed all over the China, which is wild, cheap and obtained easily. Vespae Nidus has been used in traditional medicine for a long time, as external drug commonly, and to be taken orally randomly, which is sweet, neutral in nature and in the stomach. Vespae Nidus has the function of detoxicating, destroying parasites, dispelling wind and removing pain in Chinese Pharmacopoeia(CHP). According to Great Dictionary of Chinese Medicine, it is usually been used to remove putrid, promote tissue regeneration, reduce inflammation, relieve pain and promote wound healing.So far, provisions for medicinal material base of the original, characters and clinical application are included in CHP(Edition 2010), while the projects of the identification of chemical composition and assay of Vespae Nidus have not been collected in CHP. The main component of Vespae Nidus is not clear, and the content and stability has not yet been verified. Effective control methods are needed for quality evaluation of Vespae Nidus.In view of the present research status, on the basis of the Summary of their chemistry and pharmacology, this thesis carry out a further study on chemical constituents and established several related methods for quality analysis of Vespae Nidus.Methods:Cyclohexane extracts of the Vespae Nidus were analyzed by GC and GC/MS methods. Alcohol extract were separated by solvent extraction and repeated column chromatography, The structure of separation compounds were identified on the basis of physicochemical properties and spectroscopic analysis. HPLC fingerprint and assay methods were set up further.Results: Thirteen compounds were identified on the basis of GC/MS analysis, are myristic acid(1), palmitic acid(2), heptadecanoic acid(3), stearic acid(4), oleic acid(5), linoleic acid(6), α-linolenic acid(7), arachidic acid(8), heptadecane(9), docosanoic acid(10), nonacosane(11), tetracosanoic acid(12), hentriacontane(13). A assay method of compounds 2,4, 5,6, 7 and gas chromatographic fingerprint method have been established. And two compounds were isolated and identified as: xanthurenic acid(14) and kyurenic acid(15). Among them, Compound 14-15 were isolated from this genus for the first time. The HPLC method to analysis content of two components in Vespae Nidus is simple and easy.Conclusion: This thesis reporte the identification method of the above compounds, the assay method of compound 2, 4-7, 14-15 and fingerprinting analysis of Vespae Nidus. The results will provide preliminary theoretical and experimental basis for further study and development of Vespae Nidus.
