Objective: To analyze the major genotypes of hepatitis C virus in Han, Uygur and Kazakh in one tertiary hospital in Xinjiang, and to understand the differences of the major genotypes of hepatitis C virus among the three nationalities. By reverse transcription PCR amplification and reverse dot blot hybridization technique to detect HCV genotyping and understand characteristics of the Han, Uygur and Kazakh hepatitis C patients with HCV genotype. Provide evidence for basic research, clinical treatment and disease prevention of HCV in Xinjiang. Methods: To collect department of infectious diseases, Han, Uygur and Kazakh hepatitis C serum samples in one tertiary hospital in Xinjiang, the first by fluorescence quantitative PCR detect viral load, load greater than 1x103copies/ml and then PCR reverse dot hybridization method for HCV genotyping. Results: In 669 HCV infected serum samples, PCR reverse dot hybridization is successful in 652 cases and 17 cases are not identified, the detection rate is 97.4%(652/669), 6 genotypes are 1b, 2a, 3a, 3b, 6a and hybrid 1b/2a, 1b type are 395 cases(60.58%), 2a type are 188 cases(28.83%). 1b type and 2a type’s distribution are difference between Han and Uygur, Kazakh, and the difference is statistically significance(P<0.05). 2a type in male hepatitis C infection and 1b, 2a type in women hepatitis C infection’s constitute ratio are difference between Han and Uygur, Kazakh, and the difference is statistically significance(P<0.05).In hepatitis C infection of Urumqi aera, 2a type is difference between Han and Uygur, Kazakh. And in 1b type and 2a type are difference between Han and Uygur, Kazakh in other aera except Urumqi city, and the difference is statistically significance(P<0.05). In the cases of Kazakh, 1b type and 2a type had a higher proportion, especially 2a type, the proportion of cases is higher than the Han and Uygur.1b type HCV infections have differences in the distribution of various clinical types, and the difference is statistically significance(P<0.05), in liver cancer associated with hepatitis c, proportion of 1b type is higher, at 83.33%.The average RNA loads of 1b type is(1.41×106±1.43×105) copies/ml, the average RNA loads of 2a type is(7.84×105±1.43×105)copies/ml, and the difference is statistically significance(P<0.05). Carrying capacity(1 × 104~9.99 × 104) and(1 ×107~9.99 × 107), there is significance difference between 2a and 1b type(P<0.05). Conclusions: 652 cases in Xinjiang region of hepatitis C virus(HCV) major genotype is 1b type, then 2a type, 1b and 2a type are difference between Han and Uygur, Kazakh. The ethnic groups of Xinjiang have different distribution characteristics with other cities in the north, it may be one of the reasons to lead to HCV genotype distribution and epidemiology distribution characteristic of previous reports for inconsistent. Six genotypes(1b, 2a, 3a, 3b, 6a, 1b/2a types) differences in gender, constituent ratio are no differences. While layer statisticcal by gender, the results showed that both hepatitis c infection between male and female are differences, respectively in type 2 a, 1 b, 2 a type between Han and Uygur, Kazakh. This difference may be related to three national habits, level of education, the gap between rich and poor and ethnic genotype and other factors.1b type HCV liver injury more serious than other subtypes, and there are differences in its clinical treatment and cycle therapeutic effect of interferon. To understand HCV genotyping regional distribution characteristics has important clinical and epidemiological significance.To understand Xinjiang region of HCV genotyping in patients with hepatitis c provides important information, at the same time for epidemiological studies of viral hepatitis C, judging condition and prognosis and antiviral effect provides a valuable resource.
