Objective:To analyze the relationship between HBV genotypes and HBV-DNA andmutation. Methods:The serum samples from200patients with chronic hepatitis B werecollected, HBV-DNA level was detected by the polymerase chain reaction(PCR).Genotyping of HBV was done by real-time PCR. Lamivudine and adefovirassociated resistant sites(include180L、204M、181A、236N) was analyzed by gene chips.Resμlts:(1)The distribution of HBV genotypes was not significantly different in differentgender and age(χ~2=1.818,P=0.611;χ~2=18.406,P=0.080), but showed the difference indifferent ethnicities(χ~2=89.588,P=0.000).(2)The mutational difference in the commonresistant sites were statistically significant(P<0.05).(3)Different HBV genotype haddifferent mutation rate(χ~2=33.348,P=0.000), the mutation rate of C/D mixure reached96.7%.(4)The distribution of HBV genotypes among common mutational pattern had nostatistical difference(χ~2=16.193,P=0.369).(5)The difference of HBV mutation rateamong different HBV-DNA level was statistically significant(P<0.05), but the mutationrate between middlle HBV-DNA level(105-107IU/ml) and high HBV-DNA level(≥107IU/ml) had no statistical difference. Conclusion:HBV genotypes have differentdistribution in different ethnicities;The mutational information in common resistant sites isdifferent; Differrent HBV genotypes has different incidence of resistance mutations. Thedistribution of HBV genotypes are not different in common mutational pattern. HBVmutation is correlated to HBV-DNA level, the one with high HBV-DNA level may havehigh mutation rate, but when HBV-DNA level exceeds middle level, HBV mutation is notcorrelated to the HBV-DNA level.
