Mechanism Of Acupuncture On ANP Of Adrenal In Anxiety Model

Objective:This study is supposed to observe the effects of acupuncture on ANP and NPR-A in adrenal of chronic unexpected stress anxiety rat model, Plasma ANP concentrations and Plasma CORT concentrations so as to reveal the mechanism of acupuncture on ANP in adrenal of anxiety rat.Methods:30 healthy the male SD rats were stochastically separated into 3 groups according to weight by spss17.0 statistical software, they are blank group,model group and acupuncture group, seven ways were choosed to set on Chronic Unpredictable Stress anxiety rat model, a total of 15 days,Acupuncture at "Neiguan" and "Shenmen" of one side in acupuncture group, alternately stimulating the points of the left and right side every other day, the treatment course lasted for 8 days,that’s eight frequency. Estimate the anxiety-like behavior by observing the elevated plus maze test and open-field test results. Determine the plasma ANP by RIA means,and detect the plasma CORT by ELISA, and determine ANP and NPR-A in adrenal through the immunohistochemical method.Results:1. The anxiety model activity resultsEPM detection indicators showed:model group OE% value were significantly reduced while comparing with the blank group, (P< 0.01), OT% value has the drop trend, demonstrating the modeling was successful; Compared to the model one,acupuncture group OE% value has the increased trend, the trend was similar to the blank group; indicating that acupuncture can antagonize anxiety at a certain extent.2. Plasma related concentrations(1) Plasma ANP concentrations:Compared to the blank group, ANP concentrations increased in evidence in models group(P<0.05), suggesting that secretion activity of ANP seemed excited in a certain degree in model group;When comparing with the model group,ANP concentration in acupuncture group were slightly down-regulated, and closer to the blank group,which indicated that acupuncture might down-regulate plasma ANP concentrations.(2) Plasma CORT concentrations:Compared to the blank group, CORT concentrations increased in evidence in models group(P<0.01), When comparing with the model group, Plasma CORT concentration in acupuncture group were significantly down-regulated(P<0.01), demonstrating the acupuncture can regulate the activity of the HPA axis through down-regulate plasma CORT concentrations.3.adrenal ANP relevant indicator results(1) the level of adrenal ANP:in the adrenal zona fasciculate, zona reticularis:comparing with blank group,the ANP optical density of model group slightly decreased; comparing with model group,the ANP optical density of the acupuncture group increased in evidence. demonstrating the acupuncture can acupuncture can significantly promote the expression on ANP in adrenal cortex and medulla.(2) the level of adrenal NPR-A:Comparison between groups showed no statistically significant.Conclusion:acupuncture shows antagonizing anxiety at a certain extent for chronic stress anxiety rat models, acupuncture play a role of antagonizing anxiety at a certain extent through down-regulate plasma ANP concentrations and up-regulate expression of ANP in adrenal to inhibit the release of CORT in adrenal to regulate the activity of the HPA axis.
