Experimental Research About Haoqin Qingdan Decoction Effecting The Expression Of IL-4、IFN-γ In Serum Of Mice With Chronic Gastritis After Infecting With H.Pylori

[Objective]In this research,through testing content of IL-4、IFN-γ in Serum of lab mice, we can reveal the protective effect of Haoqin Qingdan decoction to chronic gastritis of mice with spleen and stomach damp heat syndrom after infecting with H.pylori.[Method]In this research,60 SPF level C57BL/6 mice (male and female each half)were divided into 6 groups randomly,10 for each group,and separately named normal group,modeling group, li zhu wei san lian group,high-dose Haoqin Qingdan decoction group,middle-dose Haoqin Qingdan decoction group and low-dose Haoqin Qingdan decoction group.Firstly ,Except the normal group, others mice were established this model of chronic gastritis with spleen and stomach damp heat syndrom through eating fat and sweet food in the damp heat environment after infecting with H.pylori. Secondly,the normal and modeling groups mice were fed with normal saline, at the same time,the other groups mice were fed with medication accordingly,two times a day.It need 14 days in total.Finally,we dissected every mice and collected the serum and gastric mucosa.After experiment,we detected the content of IL-4、IFN-γ in the serum and observe the pathological changes of gastric mucosa.[Result](1)IL-4:Modeling group has the much higher content comparing with the normal group, the statistics are significant (P<0.01); Comparing with the modeling group,the contents are lower in li zhu wei san lian group, high-dose Haoqin Qingdan decoction group and middle-dose Haoqin Qingdan decoction group, the statistics are significant (P<0.01 or P<0.05); The content decrease obviously in li zhu wei san lian group and high-dose Haoqin Qingdan decoction group comparing with the middle-dose Haoqin Qingdan decoction group, the statistics are significant (P<0.01 or P<0.05);There is no statistical significance between the high-dose Haoqin Qingdan decoction group and li zhu wei san lian group (P>0.05)(2)IFN-y:Modeling group has the much lower IFN-ycontent comparing with the normal group, the statistics are significant (P<0.01); Comparing with the modeling group,the contents are higher in li zhu wei san lian group and high-dose Haoqin Qingdan decoction group and middle-dose Haoqin Qingdan decoction group, the statistics are significant (P<0.01 or P<0.05); Comparing with the middle-dose Haoqin Qingdan decoction group,the content increase in li zhu wei san lian group and the statistic is significant (P<0.05),but there is no statistical significance in high-dose Haoqin Qingdan decoction group;There is no statistical significance between the high-dose Haoqin Qingdan decoction group and li zhu wei san lian group (P>0.05).(3)The pathological changes of gastric mucosa:The differences are obvious on chronic and active inflammation between the modeling group and normal group (P<0.01), the degeneration and interstitial hyperemia edema are more serious in modeling group comparing with normal group which indicate successful modeling;The statistical differences are significant between the theraputic groups and normal group.The theraputic effect is the most obvious in li zhu wei san lian group and high-dose Haoqin Qingdan decoction group.[Conclusion](1)Haoqin Qingdan decoction can decrease IL-4 content and increase IFN-y content in the mice serum which has a positive correlation with the dosage of administration.(2)The Th2 is the major model on chronic superficial gastritis after infecting with H.pylori and the expression of Thl is restrained.So adjusting the unbalance is beneficial to restrain the inflammation.(3)Haoqin Qingdan decoction can improve the inflammation of lab mice gastric mucosa caused by H.pylori injection and adjust the Thl/Th2.
