Objective To explore the expression on interleukin-4 (IL-4) and interferon (IFN- γ) in stomach of Helicobacter pylori-infected subjects in Yunnan Province and investigate the expression difference on the effect of eradication therapy. Method Helicobacter pylori infection, density of helicobacter pylori and character of histo-pathology of 106 samples adapted to this study were evaluated. Immunohistochemical technique was used to determine the distribution and levels of these cytokines in the stomach. The level of IL-4 and IFN- γ secreted in culture T lymphocytes was investigated by flow cytometry before and 3 months after therapy. H. pylori infection clearance was validated by urea breath test (UBT) 1 month after therapy.Result IL-4 was found mainly in monocytes in propia lamina, while IFN-γ was mainly seen in monocytes, lymphocytes in propia lamina as well as epithelial cells. The frequency of IFN- γ positively stained MNCs in H.pylori infected patients is significantly higher than seen in un-infection subjects( z= —4.117, P<0.001) , while there was no difference of frequency of IL-4 positively stained MNCs between infected patients and infected ones (z=—1.401, P=0.161) .Moreover, in infected patients, frequency of IFN-γ positively stained MNCs in GU and CG patients is significantly higher than that in DU (z=—3.35, P=0.001; z=-2.502, P=0.012) , while there is no difference on frequency of IL-4 positively stained MNCs among GU, CG and DU patients(P>0.05). There is no correlation between the density of helicobacter pylori and IFN-γ (P=0.15) or IL-4 (P=0.23) expression, although IFN-γ expression showed inverse tendency with helicobacter pylori density .No significant...
