Effect Of Ulinastatin On Gene Expression Of Lung Tissue In Septic Rats

Abstract/Summary:
Objective: To observe the effect of ulinastatin(UTI) preconditioning on gene expression of lung tissue in septic rats by using microarray in order to study the mechanism at the genetic level. Methods: Forty-five male Wistar rats were randomly divided into control group, sepsis group and UTI group. A cecum ligation and puncture procedure was used to make sepsis model. In the sepsis/UTI group and control group, the rats were treated with intramuscular injection of balanced solution(5ml/kg) to supplement the intraoperative lost fluids and anti-shock treatment. The control group and sepsis group were treated with 1ml balanced solution an hour before cecal ligation and puncture(CLP), repeat administration every 8 hours. Whereas UTI group were treated with UTI(10×104U/kg) and administrated with appropriate balanced solution to the total volume to 1ml. Gene expression profile was studied by using RT2 Profiler? PCR Array to detect the changes in gene expression pattern of rat lung tissue after CLP. The differences of gene expression between the sepsis/UTI group and control group were analyzed by related computer software. Results: 30 differential genes were found in sepsis group compared with control group, among them 12 genes showed up-regulation and 18 genes showed down-regulation. 35 differential genes were found in UTI group compared with control group, among them 18 genes showed up-regulation and 17 genes showed down-regulation. Compared with the control group, 12 genes showed the same regulation including 4 genes with up-regulation and 8 genes with down-regulation in the UTI and sepsis group. These differentially expressed genes mainly related to inflammation, immune, apoptosis and cell growth regulation. Conclusions: Sepsis may be due to inflammation, immune response, apoptosis and cell growth regulation of abnormal gene expression. UTI has a protective effect on lung at the genetic level.
