Objective: To investigate the protective effect of ulinastatin on septic rat’s lung tissue from gene and related cell signaling pathway by gene chip technology。Methods:60 male Wistar rats were randomly divided into sham operation group,sepsis group and UTI group。septic rat model was induced by cecal ligation and puncture(CLP)in Ulinastatin group and sepsis group,but Sham operation group was experienced simulated operation without CLP.The sepsis group and the sham group were treated with intramuscular injection of saline(5ml/kg)before the operation,the Ulinastatin group were treated with intramuscular injection of ulinastatin(10*4μ/Kg,UTI10 million is dissolved in 5ml saline),Three groups were all treated with subcutaneous injection of Ringer’s Solution(10ml/Kg)after CLP,then the lung tissue of three groups were all tested by gene chip technology with RatRef-12 gene expression profile microarray after CLP.Differentially expressed genes were defined by Ratio value which is greater than 2 or less than 0.5.we used David database to classify all the differentially expressed genes and screened all the statistically significant signal pathways at the same time,then we discussed the the statistically significant signal pathways at the genetic level。Result:Compared with the sham operation group,there were 359 differentially expressed genes in the lung tissue of septic rats,of which there were up regulation of the expression of 172 and down regulation of the expression of 187.Compared with the sham group,there were 169 differentially expressed genes in the lung tissue of septic rats,of which there were up regulation of the expression of 72 and down regulation of the expression of 97.We used the method of gene enrichment pathway to screen the significant genes,we found 24 pathways in the sepsis group and 12 pathway in ulinastatin group。Two groups has 8 same pathway,but the Ulinastatin group has four signaling pathways which were not in sepsis group(ECM-receptor interaction、MicroRNAs in cancer、Glucagon signaling pathway、Non-alcoholic fatty liverdisease。)Conclusion:Ulinastatin can down regulation the inflammatory response、reduces apoptosis、alleviate hypermetabolism、 slow the progression of pulmonary fibrosis and to protect the lung tissue by regulating the gene expression level of the related pathway。...
