Objective To investigate the effects of ulinastatin on protein C(PC) and activation of protein C (APC) concentration in rats’lung tissue with severe sepsis-induced ALI/ARDS.Methods Severe sepsis induced ALI/ARDS rats were made by cecal ligation and puncture combined with normal saline lavage, taking PaO2/FiO2≤300as inclusion criteria. ALI/ARDS rats were randomly divided into ALI/ARDS group, ulinastatin50000units/Kg group, ulinastatin100000units/Kg group. Rats were sacrificed1h,2h,3h after models were completed, preparing of lung tissue homogenates after tissue weighing. Tissue PC and APC concentrations were detected by ELISA method.Results (1) Concentrations of PC:The concentrations of PC in ulinastatin100000units/Kg group [(3431+793),(3945±661),(4370±306) ng/ml] were lower than those of ulinastatin50000units/Kg group [(3826±355),(4697±389),(5289±587) ng/ml], both lower than those of ALI/ARDS group [(4324±424),(5788±409),(6008±317) ng/ml](p <0.01).(2) Concentrations of APC:The APC concentration in ulinastatin100000units/Kg group [(1341±213),(1900±338),(1802±376)ng/ml] were lower than those of ulinastatin50000units/Kg group [(1593±385),(2022±193),(1987±411)ng/ml], both lower than those of ALI/ARDS group [(3945±270),(3618±352),(2968±407)ng/ml](p <0.05).Conclusion Ulinastatin reduced PC concentration and showed a dose-dependent trend, however, ulinastatin played the role of protective effect on activation of PC.
