Objective:To investigate the effect that local infiltration of ropivacaine combined with intraperitoneal injection of sufentanil on wound healing in mice and the granulocyte-macrophage colony-stimulating factor(GM-CSF).Methods:There are 128 health of male Sprague-Dawley mice, weighting 20-25 g, according to the random number table, 128 mice were randomly divided into 4 groups(n=32): blank Group(NS group) and ropivacaine Group(L group), sufentanil Group(S group),ropivacaine with sufentanil Group(L+S group). Weight the Mice, 0.6% pentobarbital sodium were injected intraperitoneally for anesthesia. After the back hair removal,general disinfection, to make a 0.8cm×0.8cm wound in the back, cut open the skin to skin deep fascia. 10 min before incision, L group and L+S group were subcutaneous injected0.5% ropivacaine by 1.0ml/kg. L group and L+S group were intraperitoneal injected sufentanil solution 1.0ml that contains sufentanil 3.0μg/kg. Choosing 8 mice in each group at the 3, 7and 10 rd day, to check its wound healing rate after being condemned to death. Take the HE staining dye the wound tissue for the pathology detection and measure the expression of GM-CSF by the ELISA method, and use the BCA protein quantitative method to detect the total protein of all the specimens, then use the homologous ELISA and BCA value ratios represent the level of GM-CSF, and contrast the values with other groups. The 8 left mice in each group were made to check the time of complete healing.Results:(1)Wound healing rate results: In L group and S group and L+S Group, the rate of wound healing that in the 3rd, 7th and 10 th day of a manufacture wound, is respectively higher than the NS group, and L+S group is higher than the L group and S group(P<0.05). The difference of L group and S group is not statistically significant(P>0.05).(2)Pathological observation results: In the 3rd day of a manufacture mice wound,the inflammatory cell infiltration of L and S group less than the NS group. Fusiform endothelial cells and the capillaries are obviously increased. In the 7th day of manufacture wound, there are some small arteries-forming and the granulation tissue is obviously increased. In the 10 th day of manufacture wound, the small arteries are obviously increased and hair follicles occurring with the thickened epidermis layer nearly cover the wound. Compare with L and S group, the granulation tissue proliferation of the L+S group is more evident, the epithelial cells crawl earlier, the wound healing time is reduced.( 3) GM-CSF detection results: In the mice manufacture wound 3rd day, the GM-CSF level of NS group reaches a peak, the level of 7th day decreases slightly but still maintains a high level, the level of 10 th day decreases significantly. Compare with the NS group, the L group and L+S group show the same trend, the GM-CSF content of every time point is increased(P<0.05).The comparisons of the S group and NS group,the L+S group and L group, show the same trend respectively, and the differences are not statistically significant(P>0.05).(4)Wound healing time results: The wound healing of the L group is shorten than the NS group(2.000±0.000)d, the S group is shorten than the NS group(1.875±0.330)d,the L+S group is shorten than the L group and S group(2.125±0.330)d and(2.250±0.415)d respectively(P<0.05).Conclusion:The local infiltration of 0.5% ropivacaine and the intraperitoneal injection of sufentanil 3.0 μg/kg can promote wound healing, and the combination therapy is better than the alone therapy.
