Objective:To study the effect that ketorolac tromethamine combined with sufentanil on wound healing in mice.Methods:128 small mice were randomly assigned into four groups:blank physiological group(NSgroup),ketorolac group(T group), sufentanil group(S group) and ketorolac compound sufentanil(T+S group).Each mice was alone in a cage and they could eat and drink freely. After electronic scal weighing the mice,anesthesiaed the mice, under the auxiliary of hair removal device, use hair removal agent to remove the mice back hair, make the skin fully exposed.And after disinfecting, made a 1 cm× 1 cm full-thickness skin wounds on the spine midline. NS group were intraperitoneal injected 0.9% saline 1 ml, T group and S group was separately intraperitoneal injected ketorolac( the standard was 5.0mg/kg) and sufentanil( the standard was3μg/kg)which the dose was 1 ml respectively, T+S group was intraperitoneal injected both ketorolac and sufentanil which the standard was smiliar to the single drug and the composite dose was 1 ml. Eight mice in each group were executeed in the3 th,7th,10 th day to calculate the wound healing rate. And kept the 3th,7th,10 th day samples,employed the ELISIA and BCA method to calculate the amount of granulocyte-macrophage colony-stimulating factor(GM-CSF), and the specimen of three time were to make pathological section and observer the tissue change. Each group of the last remaining eight mice were calculated the wound healing time.Results:a.Wound healing time: NS group wound healing time was(15.60±0.404)days, S group was(12.35±0.314)days, T group was(12.15±0.288)days,T+S group was(10.54±0.245) days. Two single groups and compound group wound healing time were shorter than NS group wound healing time, and the compound group wound healing time was the shortest in the four groups.b.Wound healing rate:At the 3th、7th、10th day, T+S wound healing rate was the highest in the four groups,the single T group and S group were higher than the NS group, made the two single medical group to compare, there was no statistical significance(P>0.05).c. Granulocyte-macrophage colony-stimulating factor(GM-CSF): on the third day,the content of gm-csf reached its peak, on the seventh、tenth day, the content were continuously declining. The content in S group compared to the NS group,there was no statistical significance(P>0.05). The content in T + S group compared to the T group,there was no statistical significance(P>0.05).d.Pathological changes: at the 3th day, all the groups showed that inflammatory cells were infiltrating and granulation tissue were not obvious. T + S group and T group compared to NS group,the inflammatory cells were smaller. At the 7th day, all the groups showed that inflammatory cells declined, granulation tissue were still small,and the collagen were not neat.And the T + S group, epithelia cellsl expressed proliferation.At 10 th day, T group, S group, T + S group wound granulation tissue and collagen were proliferating, and the collagen were neat.Epithelial cells and fibroblasts were significantly proliferating.NS group had granulation tissue,but the collagen fiber was poorer than the other three groups.The epithelial cells and fibroblasts were also not obvious.Conclusion:Single intraperitoneal injection of ketorolac and sufentanil can accelerate wound healing in mice, and compound drugs is better than alone.But two separate drug groupto promote wound healing which is compared between group that showed no statistical significant.
