As is well-known, the content measurement of a single indicator is difficult to reflect the overall quality of Chinese medicine. As fingerprint is systematic, characteristic and reproducible and could reflect the quality of traditional Chinese medicine comprehensively, it has become an effective way of controlling the quality of Chinese medicine. In recent years, Gentiana Scraba Bge become the emphasis of Chinese medicine development, and many domestic and international experts have researched deeply about the pharmacological activity of gentian. But relative study of Rigescent Gentiana Scraba Bge have not been processed yet. Therefore, this paper mainly focused on establishing the mutual mode of Rigescent Gentiana Scraba Bge and Processing Rigescent Gentiana Scraba Bge fingerprint, which provides a basis for the evaluation of gentian quality and the standardization cultivation. The experiments established the characteristic fingerprint of Rigescent Gentiana Scraba Bge and processing Rigescent Gentiana Scraba Bge from different producing areas applying HPLC method with Gentiopicroside and Sweroside as the index component.This study committed to establish the fingerprint and its mutual mode, thus the fingerprint of Rigescent Gentiana Scraba Bge should be representative, which required in different place, time and climatic conditions. Steady HPLC method was adopted to establish fingerprint and the content of Gentiopicroside was also measured and compared with samples of different producing areas. As the similarity of fingerprint could only reflect the quality of medicinal materials qualitatively, similarity and peak area ratio was both analysed. The content of Geatiopicroside and fingerprint was combined to evaluate the quality of medicinal materials.Firstly, the optimized extraction process was selected according to the references:sealed ultrasonic extraction twice with methanol,30minutes each time. For the Processing Rigescent Gentiana Scraba Bge:Add rice wine, equal to10%of the amount of raw materials mixing embellish to yellow rice wine is absorbs, fry under150℃~200℃for10minutes.Secondly, the chromatographic conditions of fingerprint was investigated according to the guidence of <Chinese medicine chromatographic fingerprint>. There are two selections for mobile phase:methanol-water and acetonitrile-water system. Synthesizing all kinds of factors: sharp-peak and better resolution, Agilent ZORBHX SB-C18(4.6*150mm.5μm) and acetonitrile-water system was chosen. As to the selection of column temperature.Due to the column damage of high temperature.30℃condition was proved to be suitable for these experiments.According to the extraction of medicinal materials and processing technology,Rigescent Gentiana Scraba Bge and processing Rigescent Gentiana Scraba Bge sample solution were prepared. Under the condition, chromatography picture Rigescent Gentiana Scraba Bge and Processing Rigescent Gentiana Scraba Bge from10different producing areas were prepared. Chromatography S1of the similarity degree evaluating system2004of Traditional Chinese medicine promulgated by State Pharmacopocia Committee was considered as reference fingerprint. Reference Map R was produced by multi-point correction, automatic matching and the median method. According to the matching out put result there is13mutual peaks. Compared with the standard UV and retention time, peak NO.5is gentiopicroside and peak NO.6is sweroside and the peak area ratio of gentiopicroside is over40%, which was chosen as reference peak. The mutual mode of HPLC fingerprint of Rigescent Gentiana Scraba Bge and processing Rigescent Gentiana Scraba Bge was established.
