As one of the mostly influencing factors for interior quality of traditional Chinese medicines(TCM), effective constituents availably direct the clinical medication by the bioactivity. Modern medicine had shown that comprehensive evaluation of TCM was pivotal for clinic, production, preparation and market. Gentiana rigescens Franch. ex Hemsl. mainly distribute in Yunnan, Sichuan province of China is widely known as traditional Chinese herbs in folk with long medicinal history. G. rigescens(Gentianaceae: Gentiana), whose chemical constituents mainly include iridoids, triterpenes, lignans, flavanoids, alkaloids, gentisides and others, is perennial herb. Some of them have liver protection, choleretic effect, analgesia, anti-inflammatory, anti-oxidant, promoting the growth of neurons and so on. Present article concluded the chemical constituents and pharmacological activity of Gentiana rigescens to provide a reference for further developing and employing. Present study was divided into four main chapters:(Chapter One) Optimization of gentisides extraction from Gentiana rigescens Franch. ex Hemsl. by response surface methodology. On account of gentisides’ existing with other polarity components, purification and investigation to multifarious extract factors were conducted in preliminary work, and extract temperature, extract time and ratio of material to liquid were considered as most significant factors influencing extract yield. Response surface model had been successfully established and extract yield with 98.61±0.61% could be obtained.(Chapter Two) Evaluation of the Cultispecies of Gentiana rigescens and Corresponding Cultivation Techniques. We had employed LC-MS to determine the contents of iridoid glycosides by quantitative analysis of multi-components by single-marker(QAMS) method. And then we had established fast analytical method to identify the wild samples and cultural samples according to employing FT-IR combining with PLS-DA. We concluded that Gentiana rigescens cultivars was partly able to be substitute for wild species and transplant was the optimal technique along with GR artificial cultivation.(Chapter Three) Influence of Herb-Arbor Intercropping System to the Iridoid Glycosides in Gentiana rigescens. We had employed HPLC to determine the contents of iridoid glycosides and flavones in test samples from different herb-arbor intercropping system, and established fast analytical method to identify the wild samples and herb-arbor samples by using UV-vis combining with PCA and HCA. We concluded that Gentiana rigescens in Camellia sinensis intercropping system contained most gentiopicroside, sweroside and total active constituents.(Chapter Four) Influence of Material Processing to Gentiana rigescens. We had employed HPLC to determine the contents of iridoid glycosides in test samples processed with different preliminary process and used full chromatographic datas to evaluate test samples’ qualities. Analysis results revealed processing method that cleaning by rinsing and indoor seasoning was more optimizing than others.
