ObjectiveTo establish the Female BALB/c mice model of the Immunodeficiency with Pulmonary infection, for detecting the effects of traditional Chinese medicine Qingfeipeiyuan Soluble Granules(QSG) on the T lymphocyte subpopulation, IFN-γ,IL-1β,IL-6,IL-10 and TNF-α with Immunodeficiency and Pulmonary infection by mouse models, Analyzing the characteristics of QSG and efficacy.MethodsDividing 72 female BALB/c mice into 6 groups randomly,such as Normal group, Model group(Immunodeficiency and Pulmonary infection group), Western medicine group(AZT+PPT group), Tangcao tablet group, QSG high dose group, QSG low dose group. In addition to the normal group, all of the groups inoculate the virus of the FLV by intraregional injection to establish the model of Immunodeficiency. 21 days later, dripping the streptococcus pneumoniae into nasal cavity, then the mouse model of Immunodeficiency and Pulmonary infection is established. Each treatment group began to treat respectively. The percentage of the T lymphocyte subpopulation in blood serum including the CD4 T lymphocyte and CD8 T lymphocyte were detected by Flow Cytometry;The expression of IFN-γ,IL-1β,IL-6,IL-10 and TNF-α of the lung was detected by Immunohistochemistry; The morphology of the lung tissue pathology of the mouse was observed under electron microscopy.Result1 The effecting of General results in the complex model mouse: Compared with normal group, weight loss,poor hair gloss, significantly reduced diet,activity gradually reduced, but the improvement in each treatment group.2 The function in thymus mass and spleen mass of the complex model mouse: Compared with normal group, model group thymus mass were lower than the normal group, the difference was statistically significant(P<0.01);Compared with model group, QSG high dose group can increase the degree of thymus mass, the difference was significant statistically(P <0.05). Compared with normal group, model group spleen mass were higher than the normal group, the difference was statistically significant(P<0.01);Compared with model group, Except QSG low dose group, the other treatment groups can reduce the degree of spleen mass, the difference was significant statistically(P <0.05).3 The function in thymus index and spleen index of the complex model mouse: Compared with normal group, model group thymus index were lower than the normal group the difference was statistically significant(P<0.01);Compared with model group, except QSG low dose group and Tangcao tablet group, the other treatment groups can increase the degree of thymus index, the difference was significant statistically(P <0.05). Compared with normal group, model group spleen index were higher than the normal group the difference was statistically significant(P<0.01);Compared with model group, Except QSG low dose group, the other treatment groups can reduce the degree of spleen index, the difference was significant statistically(P <0.05).4 The effecting of T lymphocyte subpopulation with the complex model mouse: Compared with model group, except QSG low dose group, the other treatment groups can increase the percentage of CD4 T lymphocyte subpopulation and the ratio of CD4/CD8, Western medicine group increase the ratio of CD4/CD8 was higher than the other treatment groups, the difference was significant(P <0.01), the others difference was significant statistically(P <0.05). Compared with model group, except QSG low dose group, the other treatment groups can reduce the percentage of CD8 T lymphocyte subpopulation, Western medicine group reduce the percentage of CD8 T lymphocyte subpopulation was lower than the other treatment groups, the difference was significant(P <0.01), the others difference was significant statistically(P <0.05).5 The effecting of IFN-γ,IL-1β,IL-6,IL-10,TNF-α with the complex model mouse: Compared with model group, except QSG low dose group, the other treatment groups can reduce the expression of IL-6,the difference was significant statistically(P <0.05). Compared with model group, Except QSG low dose group and Tangcao tablet group, the other treatment groups can reduce the expression of IFN-γ,IL-1β,TNF-α, the difference was significant statistically(P <0.05). Compared with model group, Except QSG low dose group and Western medicine group, the other treatment groups can increase the expression of IL-10,the difference was significant statistically(P <0.05).6 The morphology of the complex model mouse: Lung tissue of model group associated with the inflammatory response, the result show that the epithelial cells of the bronchioles has cell degeneration and necrosis 、 cilia lodging; Lung interstitiums increased; A large number of lymphocyte,neutrophil, and macrophages exudation. Compared with model group, The morphology of all treatment groups is change lightly, and the improvement in Western medicine group QSG high dose group.ConclusionThe complex model of Immunodeficiency with Pulmonary infection is established successfulNot only QSG can increase thymus mass、thymus index, the percentage of CD4 T lymphocyte subpopulation and CD4/CD8, but also can induce spleen mass,spleen index and the percentage of CD8 T lymphocyte subpopulation, It’s show that this medicine can regulate the immunity function.Not only QSG can effectively increase the expression of IL-10,but reduce the expression of IFN-γ,IL-1β,IL-6,TNF-α. We can see that QSG can control bacteria.
