This study was systematically condected to investigate the effects of Chenpi (Pericarpium Citri Reticulatae) on Baizhu Shaoyao San chemical compositions and efficacy, to explain the compatibility mechanism of Baizhu Shaoyao San in Chenpi and provides scientific basis and experimental foundation for the further research on traditional Chinese medicine compatibility mechanism in the future exploration, and establish a the new study method.By HPLC-DAD method for the simultaneous determination of 10 marker compounds, compared and analyzed the content changes of 10 marker compounds after the combination of Chenpi. The result showed that Chenpi compatibility can make the amoumts of albiflorin, narirutin, 4’-O-β-D-glucosyl-5-O-methylvisamminol, hesperidin and nobiletin were significantly raised. In the meantime, Q-TOF-MS was employed to qualitatively and quantitatively compare the influence on the main compositions caused by Chenpi compatibility, constituents in the positive mode and negative mode were recognized that have relatively large differences.The volatile components in Baizhu Shaoyao San was analyzed qualitatively by using GC-MS, the results indicated that volatile compounds have the obvious change in Baizhu Shaoyao San after Chenpi compatibility, and certain chemical ingredients appeared or disappeared at the same time. However, the majorty different components were come from Chenpi completely.The trace element samples were prepared with microwave assisted digestion, and determined by ICP-AES. The method was successfully validated and it was used to simultaneously determine seven important trace elements. The result shows that Baizhu Shaoyao San has a variety of trace elements, and it might be one of the reasons why Baizhu Shaoyao San can have great use for IBS and other diarrhea illness. The another result shows that the solubility of trace elements, especially Fe, Mg, Zn,Cu, have a definitely difference after Chenpi compatibility.Make a research on the content of polysaccharide in Baizhu Shaoyao San and then compared the differences before and after Chenpi compatibility. Results showed that Chenpi compatibility can made polysaccharide components increase.Study on the growth promoting effect of Baizhu Shaoyao San of Bifidobacterium in vitro, and compared the effects of Chenpi before and after compatibility, found that growth promoting effect enhanced obviously after Chenpi compatibility.By observating the Baizhu Shaoyao San on general physiological index of diarrhea-irritable bowel syndrome model mice, investigated the pharmacodynamics of Baizhu Shaoyao San on diarrhea-irritable bowel syndrome, then comparedthe effect before and after Chenpi compatibility, found that Without-Chenpi Baizhu Shaoyao San medicine effect significantly weaker than the original one.
