This study was systematically condected to investigate the influence on chemical constituents, pharmacokinetics and efficacy of Baizhu Shaoyao San maded by Chinese herbal medicine processing, providing some scientific basis and studying accordance for comprehensively evaluating the effect on the prescriptions caused by processing, then integratedly studying on the scientific connotation of processing and prescriptions, and establishing new research ideas and modes.On the basis of the optimization about the extraction process of non-volatile constituents in Baizhu Shaoyao San by single factor and orthogonal design experiments, HPLC-DAD was carried out to analyze the changes in the amounts of7chemical components in Baizhu Shaoyao San before and after processed, the result showed that the amounts of gallic acid, albiflorin, narirutin, hesperidin, and atractylenolide I were significantly raised after processed, while the amount of nobiletin was distinctly decreased; In the meantime, UFLC/Q-TOF-MS was employed to qualitatively and quantitatively compare the influence on the main compositions caused by processing,41constituents in the positive mode and31constituents in the negative mode were recognized as compositions that have relatively large differences.GC-MS was successfully developed to determine the volatile constituents in Baizhu Shaoyao San before and after processed. The results indicated that after processed, the total content of volatile oil in Baizhu Shaoyao San was significantly reduced, the relative content of most components tended to be decreased, and certain chemical ingredients appeared or disappeared at the same time.A rapid, sensitive and specific liquid chromatography/tandem massspectrometry method (LC-MS/MS) was developed for simultaneous determination of3active components (paeoniflorin, narirutin, hesperidin) in rat plasma, the difference of the3constituents between non-processed and processed Baizhu Shaoyao San on the pharmacokinetics was compared. The results showed that the Cmax, AUC (o-8h), AUC (0-∞) and MRT of the3constituents in rat plasma were conspicuously increased, indicating that Baizhu Shaoyao San can promote the absorption, prolong mean retention time and improve the bioavailability of active constituents in rat after processed.The influence on the efficacy in treating diarrhea-predominant irritable bowel syndrome by Baizhu Shaoyao San before and after processed was inspected by observing the general physical condition, intestinal motility and measuring the amount of relevant indicators SP and VIP in plasma of the diarrhea-predominant irritable bowel syndrome model mice. The results showed that the anti diarrhea effect of Baizhu Shaoyao San was enhanced after processed, the mechanism may relate to the inhibition of intestinal movement and the decrease of some corresponding brain-gut peptidein in the plasma of the model mice.
