Objective:Through the observation of the effect of electroacupuncture(EA) treatment on expression of phosphatase and tensinhomology deleted on chromosome ten (PTEN) and growth associated protein-43 (GAP-43) protein, to investigate the possible mechanism of Electroacupuncture on neural regeneration and functional recovery after ischemia.Methods:24 healthy adult male SD rats were randomly divided into sham group, focal cerebral ischemia model group and EA group with 8 in each group.At the third day after the models were established, the rats of EA group accept treatment of EA at Baihui and fengfu Acupoints, daily maintenance 30mins. The model group was fixed only, sham group did not accept the deal.We observe the behavior of rats with corner Test and detect the expression of GAP-43 and PTEN protein around infarct zone with Western blot method after 14 days.Results:After 14 days, the number of turn right of EA group was significantly decreased compared with model group (P<0.05); At the day before operation, the number of turn right and left of sham group and model group is no difference. At the third day after the models were established, the number of turn right of EA group and model group was significantly decreased compared with sham group (P<0.05).At 7 days after treatment, the chance of turn right of EA group and model group was descended compared with at the third day (P>0.05) The chance of turn left and right of sham group is no difference all the time.The expression of GAP-43 protein in rat of model group was increased compared with the sham group (P<0.05), and increased significantly in the EA group than in the model group and sham group (P<0.05). The PTEN expression have no statistical significance between the model group and the sham group (P>0.05), but there is a downward trend in model group. While the expression of PTEN protein in the EA group were significantly decreased than those in the model group and sham group rats (P<0.05).Conclusion:EA may promotes nerve regeneration and functional recovery after ischemia by inhibiting expression of PTEN protein and increaseing expression of GAP-43 protein which at semidarkness.
