Objective To study the effects of electroacupuncture on differenraition of endogenous nerve stem cells in rats with cerebral ischemia and to probe the possible mechanism of acupuncture curing stroke.Methods Made the models of rats with the middle cerebral artery blocked, then randomly divided into two groups:the electroacupuncture group and the model group. The electroacupuncture group were stimulated the acupoints of "Bai Hui" and "Feng Fu" by electroacupuncture. The model group were not electroacupunctured. Every rat was given 5-bromodeoxyuridine by intraperitoneal injection. Reperfusion by saline and paraformaldehyde, the materials drawn, fixed by paraformaldehyde, general dehydration and transparency by gradient alcohol and xylene, dipped wax, embedded, coronal slices. The immunofluorescence double labeling of the 5-bromodeoxyuridine/neun-specif icenolase(BrdU/NSE)and 5-bromodeoxyuridine/glial fibrillary acidic protein(BrdU/GFAP) were done. To count the quantity of the positive expression cells of BrdU/NSE and BrdU/GFAP under the immunofluorescence microscopy. The experimental data were analyzed by statistical software of the SPSS13.0. The factorial design analysis of variance was used between the two groups. The completely randomized design analysis of variance was used in one group.Results 1. The expression of the positive cells of BrdU/NSE:In contrast, it was significantly different between the two groups (F=1069.23, P< 0.01). It was also significantly different for the interaction between the two groups (F=74.156, P< 0.01). Compared to different times in one group, it was significantly different in the model group (F=1.90, P<0.01), it was significantly different in the electroacupuncture group (F=355.51, P<0.01); 2. The expression of the positive cells of BrdU/GFAP:In contrast, it was significantly different between the two groups (F=28100.21, P< 0.01). It was also significantly different for the interaction between the two groups (F=371.258, P< 0.01). Compared to different times in one group, it was significantly different in the model group (F=6.50, P< 0.01), it was significantly different in the electroacupuncture group (F=90.18, P< 0.01)Conclusion Electroacupuncture could promote the expression of NSE and GFAPof rats with cerebral ischemia. It indicated that acupuncture could promote the endogenous nerve stem cells in rats with cerebral ischemia to differentiate into neurons and neural glial cells.
