Background Using the GWAS method to study a large number of Han Chinese severe acne samples, preliminary studies showed that the SELL gene is the susceptible gene of severe acne. But the correlation between SELL gene and severe acne has not been reported at the molecular level, and still need to do further research,the research is in this direction.Objective To investigate the difference in molecular expression level of gene SELL between severe acne and healthy control.Methods 1) Collected 86 cases of HanChinese severe acne and 100 healthy controls as the object of study by Pillsbury classification method, the clinical data gained by questionnaire and filling out the unified design of the questionnaire and the health survey.2)Total RNA was extracted from peripheral white blood cells, one by one, detection with the expression level of SELL gene mRNA of peripheral blood leukocytes in patients and controls by real-time quantitative PCR method, using 2(-△△CT) (delta delta CT) method to obtain the results by beta-actin in reference genes.3) Peripheral serum was separated, one by one, using ELISA method to detect serum soluble L-selectin concentration of heavy acne and healthy control.4)The data was analysised by using independent samples t test analysis, chi square test and two classification Logistic regression.Result 1) L-selectin mRNA of severe acne and healthy control reverse transcriptase expression level observably difference, and the difference was statistically significant (β=0.534, OR=0.238, P<0.05,95%CI=1.07-2.27)2) Serum soluble L-selectin of acne and healthy control significantly divergence (β=0.05, OR=0.22, P<0.05,95% CI=1.01-1.10)Conclution SELL gene mRNA reverse transcriptase and serum soluble L-selectin levels of severe acne compared with healthy controls was higher, suggesting that upregulation of the expression of SELL gene in severe acne of the level of transcription and translation, revealing the expression of SELL gene in severe acne on the molecular level. SELL gene with severe acne has relativity on molecular expression.
