Researching On Haploid Type Of The Androgen Receptor Gene Of Severe Acne In Chinese Han Nationality In Yunnan Province

Objective:It is reported that androgen plays a crucial role in the occurrence, development and continuous state of the severe acne. The androgen receptor polymorphism loci which are encoded by AR gene were selected to study the correlation between the genotype haploid and severe acne. This would reveal the genetic pathogenesis of severe acne, which in turn provides the theoretic basis for the genetic research of severe acne.Patients and Methods:Two hundred eleven healthy individuals(male95, female116, age27.5±7.9years old) and Two hundred eighty-eight patients with severe acne (PillsburyⅢ-Ⅳ, male87cases, female201cases; age23.4±6.4years old) in Han nationality in Yunnan province were included in the study. Genomic DNA was extracted from peripheral blood of all patients and controls. The CAG and GGC repeat regions were amplified by polymerase chain reaction (PCR) with fluorescence-labeled primers. The microsatellite study on the CAG, GGC of AR gene was conducted by PCR-GeneScan analysis. The correlation between the polymorphism in genotype haploid of AR and increased risks suffering from severe acne were analyzed by Statistical analysis with SPSS17.0.Results:In Han nationality, The number of CAG repeats mainly distributed in18to20, The number of GGC repeats mainly distributed on15. In female, The number of CAG repeat equal23or24had a statistically significant risk suffering from severe acne compared with others (P=0.016, OR2.652(95%CI)=(1.2-5.865); P=0.011, OR3.717(95%CI)=(1.356-10.187)]. In male, The number of GGC repeat equal15had a statistically significant risk suffering from severe acne compared with others (P=0.019, OR2.167(95%CI)=(1.07-4.14); the haplotypes CAG>18/GGC≤14, or CAG≤20/GGC>14compared with CAG>20/GGC>14had statistically significant risk suffering from severe acne.(P=0.012, P=0.002). Conclusion:We speculate that (1) In the female of Han nationality, the number of CAG repeat equal23or24would have a risk suffering from severe acne.(2)In the male of Han nationality, the number of GGC repeat equal15would have a risk suffering from severe acne.(3)In the male of Han nationality, CAG>20/GGC≤14、 CAG≤20/GGC>14would be one of increased risk factors suffering from severe acne.
