Background and ObjectiveAs we all know, gastric cancer is a serious disease in our county. An early diagnosis and treatment is the key to gastric cancer prevention and control. At present, early diagnosis of gastric cancer depend on endoscopy, but because of the high cost and low patients acceptance, endoscopy cannot widely be used for early diagnosis. We don’t have a useful blood molecular marker. Our previous experiment shows that the peripheral bloodmicroRNA-23a has discrepant expression between gastric cancer group and non-gastric cancer group. This study is aim to collect peripheral blood from different patients, analyzing the expression of miR-23a level, and to find the relationship between miR-23a level and gastric cancer, and its correlation with pathological and clinical significance. Proving weather miR-23acan bea novel molecular marker for diagnosis of gastric carcinoma andcancerization monitoring.MethodPeripheral blood samples were getting from the patients of Sir Run Run Shaw Hospital affiliated Zhejiang University School of Medicine, theclinical data also collected. Blood serum was collected by centrifugation. After that, mirVanaTM PARISTM Kit was used to extract total RNA from the serum, and All-in-OneTM miRNAqRT-PCR was used to exam the miR-23a expression level. The experimental data and the clinical data were combined to analyze the results with using t-test, Chi-square test and spearman’s correlation.Result1)the expression of miR-23a in peripheral blood in gastric cancer group was significantly higher than that in healthy control group.2) when choosing CT<12.335as a positive boundary value, the true positive rate is63%, the true negative rate is60%. In gastric cancer group, the rate oflymphatic metastasis is significantly higher in miR-23a positive patients than negative patients.3) In gastric cancer group, the peripheral blood expression level of miR-23a and signet-ring cancer were positively correlated, the peripheral blood expression level of miR-23a and differentiation were negatively correlated.ConclusionMiR-23a level in peripheral blood as a molecular marker for gastric cancer diagnosis, may be helpful to judge the prognosis of gastric cancer patients.
