Objective:To reaserch the expression changes of peripheral blood miRNA-378in gastriccancer and non-gastric cancer lesions(pre-gastric cancer and healthy controls), andto explore the significance between the expression of miR-378changes and clinicopa-thologic factors.Methods:Collect20gastric cancer patients,10pre-gastric cancer patients and20case ofnormal healthy peripheral blood specimens and clinicopathological data.we usedqRT-PCR to detect the expression level of miRNA-378in the serum of peripheralblood specimens.According to the clinicopathological data of the patients, compar-ison between patients with gastric cancer and pre-gastric cancer patients in the serumof peripheral blood of normal controls expression of miRNA-378differences. Accor-ding to the Ct value tested by qRT-PCR while use the relative quantitative method2△Ctto calculate the expression of miRNA-378.The expression fold change valuesconverted to Log2△Ct,while the△Ct=CtmiRNA-378-CtU6of miRNA-378and analysisthe relationship between expression and clinicopathologic factors.Results:1) Small amount of miRNA-378was detected in peripheral blood using aqRT-PCR platform.2)The average expression level of miRNA-378in peripheral blood of gastriccancer group(-0.859±1.764)(Log2△Ct)was significantly higher than that inpre-gastric cancer group(0.882±0.966)(p<0.05) and that in healthy control group(1.646±1.54)(p<0.05).Also the average expression level of miRNA-378inpre-gastric cancer group was significantly higher than that in healthy (p<0.05);3)Through history, clinical data, experimental results, analysis of patients withgastric cancer, peripheral blood of patients with gastric cancer expression level ofmiR-378was not correlated with gender,age,tumor stage,tumor type and tumordifferentiation of infiltrative metastasis (P>0.05). Conclusion:1) Small amount of miRNA-378was detected in peripheral blood using aqRT-PCR platform.2) Peripheral blood of patients with gastric cancer expression level of miR-378was not correlated with gender,age, tumor stage,tumor type and tumor differentiationof infiltrative metastasis (P>0.05).3) Peripheral blood of miR-378is expected to become a new kind of biologicalindicators of gastric cancer diagnosis and screening。...
