The Clinical Significance Of Plasma Exosomal B7-H3,PD-L1,MicroRNA-29a,b,c And MicroRNA-570 Levels In Gastric Cancer | | Posted on:2019-07-30 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Wang | Full Text:PDF | | GTID:2404330545473426 | Subject:Clinical medicine | | Abstract/Summary: | PDF Full Text Request | | Part 1 The Clinical Significance of Plasma Exosomal B7-H3 and PD-L1 in Gastric Cancer Objective: To preliminarily study the clinical significance of plasma exosomal B7-H3 and PD-L1 levels in gastric cancer.Methods: The study including 65 cases of gastric adenocarcinoma and 18 cases of chronic superficial gastritis as controls from June 2017 to September 2017.The plasma exosomal B7-H3 and PD-L1 levels were determined by enzyme-linked immunosorbent assay(ELISA).The relationship between the plasma exosomal B7-H3,PD-L1 levels and the clinicopathological features of gastric cancer was preliminarily analysed,and the diagnosis values of plasma exosomal B7-H3 and PD-L1 levels for gastric cancer were preliminarily evaluated.Results: The plasma exosomal B7-H3 levels were significantly higher in gastric cancer than in controls(P=0.021).The plasma exosomal PD-L1 levels had no difference between gastric cancer and controls(P>0.05).Plasma exosomal B7-H3 was positively correlated with PD-L1(r=0.208,P=0.032).There was no significant correlation between the plasma exosomal B7-H3 levels and the clinicopathological features of gastric cancer(P > 0.05).The plasma exosomal PD-L1 levels were associated with the infiltration depth and the TNM stage to gastric cancer(P = 0.033,P = 0.004).The diagnosis value of plasma exosomal B7-H3 for the stage I gastric cancer(AUC=0.712)was higher than that for total gastric cancer(AUC=0.679).Conclusions: The Plasma exosomal B7-H3 and PD-L1 levels were related to the progression of gastric cancer.The plasma exosomal B7-H3 can be used as a potential marker for diagnosis of gastric cancer,especially in the diagnosis of the stage I gastric cancer.The plasma exosomal PD-L1 may play an important role in gastric cancer invasion and progression.Part 2 A Clinical Study on The Plasma Exosomal MicroRNA-29 a,b,c and MicroRNA-570 Levels in Gastric CancerObjective: To preliminarily study the clinical significance of plasma exosomal MicroRNA-29 a,b,c and MicroRNA-570 levels in gastric cancer.Methods: The study including 65 cases of gastric adenocarcinoma and 18 cases of chronic superficial gastritis as controls from June 2017 to September 2017.The plasma exosomal MicroRNA-29 a,b,c and MicroRNA-570 levels were determined by real-time quantitative polymerase chain reaction detecting system(q PCR).The relationship between the plasma exosomal MicroRNA-29 a,b,c,MicroRNA-570 levels and the clinicopathological features of gastric cancer were preliminarily analysed,and the diagnosis value of the above 4 plasma exosomal MicroRNAs for gastric cancer were preliminarily evaluated.Results: The plasma exosomal MicroRNA-29 a,b,c levels were significantly lower in gastric cancer than in controls(P=0.001,P=0.001,P=0.001),while the plasma exosomal MicroRNA-570 levels were notably higher in gastric cancer than in controls(P < 0.001).The diagnosis values of the plasma exosomal MicroRNA-29 a,b,c and MicroRNA-570 for the stage I gastric cancer were higher than that for the total gastric cancer.The plasma exosomal levels of the above 4 plasma exosomal MicroRNAs were positively correlated with the differentiation degree,the distant metastasis,the infiltration depth,the lymph node metastasis and the TNM stage to gastric cancer(P < 0.05).Conclusions: The plasma exosomal MicroRNA-29 a,b,c and MicroRNA-570 were associated with the progression of gastric cancer.All of the 4 plasma exosomal MicroRNAs can be used as potential markers for gastric cancer,especially for the stage I gastric cancer. | | Keywords/Search Tags: | exosome, gastric cancer, B7-H3, PD-L1, MicroRNA-29, MicroRNA-570 | PDF Full Text Request | Related items |
