| Purpose:Though the experiment on the Xiayuxue Tang with the "Wind Herbs" ginger and Gentiana macrophylla and the simple Xiayuxue on liver fibrosis in rats, the comparasion was made on the index of the rat liver fibrosis before and after the treatment, to study the mechanism of the wind herbs’synergistic effect, to provide a theoretical basis for the "Wind herbs" synergy effect in promoting blood circulation to resolve blood stasis, so to guide the clinical use.Methods:84rats, weight200g±20g, clean grade, Wistar male were fed for one week for adaptation, then, seven of which were randomly selected to be in the control group (A), and the remaining77rats were tentatively made modules. There was no any treatment in the control group (A), only with conventional feed. Modules were madetentative liver fibrosis models by concentration of50%CCl4olive oil solution,4ml subcutaneously injection, twice a week, a total of eight weeks.Week8, randomly selected two in the experiment group and one in the control group.Remove the liver of rats killed:①Observed the liver surface texture, size, edge, and color with the naked eye②In microscope, the pathological hepatic lobules, the liver cells arranged in cords, hepatocyte swelling degree in order to determine whether the modeling succeeded. Then the, the model group were randomly divided into six groups, i.e. model group (B), Xiayuxue tang (C), low-dose ginger compatibility group (D), high-dose ginger compatibility group (E), Gentiana low-dose comptibility group (F), Gentiana high-dose group (G), Each group had12rats.Week9, In the control group (A) and model group (B) normal saline was filled4ml/per time once a day for four weeks. Other groups (C、D、E、F) were administered with the corresponding drugs,1day once, each time4ml for4weeks (approximately8times of clinical usage for an65kg adult).12weeks later, all rats were killed:①femoral vein blood, to check serum fibrosis markers HPC Ⅲ, Col Ⅵ, LN content.②liver resection, partial liver tissue were made into liver tissue biopsy. Observing the organizational changes of lobular structure, cell cord arrangement and the extent of liver cells swelling, the vessel wall and hepatic portal area.③in the remaining liver tissue homogenates, to detect oxidation-antioxidant index MDA and SOD, liver fibrosis index Hyp, indicators of liver fibrosis factor PDGF, TGF-β1and CTGF.Results:Comparasion between "Wind herbs" Xiaxueyu Tang group, simple Xiayuxue Tang group and model control group:①the rat liver fibrosis, steatosis and liver cells edema were improved to varying extent,②in tissues fibrosis index factors TGF-β1, PDGF, CTGF reduced; oxidation-antioxidant index MDA reduced, SOD increased.③The lliver fibrosis index Hyp reduced.④in serum biochemical markers LN, HPC-Ⅲ and Col Ⅵ of liver fibrosis concentration reduced to varying extent⑤Xiayuxue Tang with "wind drug" group can resist rat liver fibrosis, steatosis and liver cell edema, better than the simple Xiayuxue Tang.Conclusions:1."Wind herb" Ginger and Gentiana compatibile with Xiayuxue Tang has stronger action in improving rat liver fibrosis, steatosis and liver cell edema than simply Xiayuxue Tang. Thus,"Wind drugs" have a synergistic effect for bettering blood circulate to resolve stasis.2. Wind drug’s synergistic effect targeting point may be associated with the changes of the release of the liver tissue factors TGF-β1, CTGF, while the tisuss’ antioxidant capacity is increased.3. The mechanism of the "Wind drug"’s synergistic effect for blood circulation to help resolve blood stasis is connected with its such natures as pungent, with light and thin quality-including ascending, scattering, penetrating, inroading, opening up, drying and acting, which can disperse evils and go up and down and clear out the accumulated masses, regulate qi’s flowing, guide the herbal tropism and lifting all the drug’s action.Therefore,"the wind drugs" compatibile with the drugs which can activate blood to resolve blood stasis has the synergistic effect. |
