Objective:Osteoarthritis (OA) is the most common of the musculoskeletal disease. It is characterized structurally byarticular cartilage degradation. The pathophysiology of the process by which whole joint degeneration ofosteoarthritis remains poorly understood. The disease affects millions of individuals aged45years andabove, and is increasing in frequency and severity within the context of the ageing population. In recentyears, with the development of biomedical technology, more and more domestic and foreign scholars arebeginning to focus on the relationship between the family of matrix metalloproteinases (MMPs) and OA inorder to clarify the pathogenesis of OA. In this experiment, we studied the expression and correlation ofMMP14/-9at the level of gene in the patients’ synovial tissues with OA and non-OA, to explore the role ofMMP-14/-9in the development of OA.Methods:68surgically removed synoviums were obtained from the patients with consent in the department oforthopedics of the first affiliated hospital of Shi He-Zi University from the Nov,2011to the August,2013,which operation include the knee joint of arthroscopy, arthroplasty, amputation et al. All of the patientswere diagnosed by the doctor of orthopedics combine with clinical symptoms and imaging (According tothe American Rheumatic Association (ARA)1995revised criteria for OA).The synovium were washedthree times with saline in one hours after surgery, quick-freezed in liquid nitrogen and stored in-80℃refrigerator for experiment. Finally, Trizol reagent to extracted RNA and semi-quantitative RT-PCR todetected the expression of MMP-14mRNA and MMP-9mRNA in68cases of synovium. The results ofmeasurement data adopt mean±standard deviation(±s), the statistical significance was determined byStudent’s t-test and Pearson correlation analysis using the SPSS17.0program, P value less than0.05wereconsidered to be statistically significant.Results:We observed the expression of MMP-14mRNA and MMP-9mRNA in all of patients’ synovium (OA andNon-OA patients); and the expression of MMP-14/MMP-9mRNA were significantly higher in OA group(0.9887±0.075/0.742±0.069) compared with Non-OA group (0.244±0.0614/0.176±0.05,P<0.05); Pearsoncorrelation analysis indicates that the expression of MMP-14and MMP-9genes were positively correlatedwith OA synovium (r=0.863,P<0.05).Conclusion:The expression of MMP-14mRNA and MMP-9mRNA was significantly higher in OA group synovialtissue compared with Non-OA group, and positively correlated with each other in OA synovium. Theresults indicated that MMP-14and MMP-9involve in the pathogenesis of OA; and to a certain extentsynergy to promote the progression of osteoarthritis.
