Objective:Intracranial arterial thrombosis, bleeding occurs which affects the normal functioningof regional cerebral blood supply organizations, this disease is seriously affecting thehealth of our population is the stroke, the main cause is atherosclerosis of the arteries, coronary heart disease, hypertension, previous family history of disease, followed byfactors such as high blood cholesterol can lead to stroke. Now after a range ofepidemiological investigations revealed that the stroke is often not caused by single genes, living environment and genetic interaction, the occurrence of diseases. So the way to betterprevention and treatment of stroke, explore the role of genetic mechanism is verypromising. This selected set of principles is based on the type of stroke, using geneexpression microarray analysis, filter out different types of familial aggregation of cerebralapoplexy and platelet adhesion system highly relevant gene SNP, on this basis, usingreal-time quantitative PCR technique method of verifying platelet adhesion systems SNPgene promoter alter gene transcription. Finally arrived at about the different types of themolecular genetics of familial aggregation mechanism of stroke, become a strong basis forindividualized stroke prevention. Methods:Experiment select has December2012to December2013in Tarzan medical schoolsubsidiary hospital neural medicine hospital of,10name family in the of brain deficiencyblood patients (including first levy who), over the family patients brain stroke in the totalof75%;10name family midbrain bleeding patients (including first levy who), over thefamily brain stroke in the patients total of75%; also select6name over in this homeshealth body no brain vascular disease family history of normal who as controlled group, controlled group and experiment group with national, men and women gender proportionbalanced, Difference of no more than5years of age, and in the same age group. Cases anda total of26cases of the control group. Using unified of investigation table on selected Home Department of first card who to and relatives for investigation, records all researchobject of General clinical information, in consent of all client object agreed Hou forsampling, used gene expression spectrum chip analysis, filter out respectively and braindeficiency blood family gathered sexual and brain bleeding of family gathered sexualheight related platelet stick attached system gene SNP, this results based Shang, usesreal-time quantitative PCR technology method confirmed platelet stick attached systemgene of started child SNP change related gene of transcription activity. Draw on differenttypes of the molecular genetics of familial aggregation mechanism of stroke, become astrong basis for individualized stroke prevention.Results:1. the Group of Group of cerebral hemorrhage and cerebral ischemia in age, smokinghistory, drinking history, history of diabetes mellitus, systolic blood pressure, diastolicblood pressure, fasting blood glucose and serum lipid level in comparison with the controlgroup, statistically significant differences were found (P>0.05).2. the human genome in peripheral blood and platelets of different types ofstroke-related genes include: GP VI, CD63, CD59, CD36, GPⅰb alpha, GPII b, GPIII a,PDGF, and vWF, and TSP, PECAM-1, ICAM-1, VCAM-1, FP4, PAF, and CD62p, TxB2,6-K-PGF1a, ITGB3.3. platelet PF4and real-time quantitative PCR method validation results: FP4in agroup reduced expression of familial aggregation of cerebral ischemia, a statisticallysignificant difference (P<0.05); group also reduced expression of PF4genes inintracerebral hemorrhage, the difference was not statistically significant (P<0.05).4. platelet CD36gene and real-time quantitative PCR method validation results:CD36gene in a group reduced expression of familial aggregation of cerebral ischemia, astatistically significant difference (P <0.05); Group also reduced expression of CD36genein cerebral hemorrhage, a statistically significant difference (P <0.05).5. the platelet ITGB3gene and real-time quantitative PCR method validation results:ITGB3gene in a group expression of familial aggregation of cerebral ischemia decrease, the difference was not statistically significant (P>0.05); group expression after cerebralhemorrhage is reduced, the difference was not statistically significant (P>0.05).Conclusion:1. The gene PF4and CD36may had relationship with the pathogenesis of IschemicStroke.2. The PF4gene and CD36gene in familial aggregation of cerebral hemorrhage with reduced expression of the group, speculated that PF4and CD36gene family aggregation ofcerebral hemorrhage may be a risk factor.3. Group ITGB3gene in familial aggregation of cerebral ischemia and intracerebralhemorrhage group expression is reduced, but the difference was not statistically significant, suggesting ITGB3gene expression is not low familial risk factors of cerebral hemorrhageand cerebral ischemia.
