Purpose: To observe the nNOS positive neurons expressing in monocular visiondeprivation amblyopia rat visual cortex and lateral geniculate body and retinal of acupunctureat the sensitive period of visual development.Discuss its role in the pathogenesis of formdeprivation amblyopiaf,summarize the visual cortex developmental status before and afteracupuncture intervention effect law, in order to clarify the mechanism of acupuncturetreatment of amblyopia provide new ideas, to find a method of treatment of amblyopia clinicalreference.Method:36wistar rats only2-week-old, using random number table method is dividedinto three groups, this is normal group, the model group, acupuncture group,cut off themodel group and the acupuncture group of rats’ lid margins, and suture experiment eye, makethem form monocular visual deprivation rat; Normal rats don’t do anything and normally bred.Model rats only make mode and not acupuncture; acupuncture group give acupuncturerespectively at the model reproduction after7day, to select four acupoints Jingming、Cuanzhu、 Guangming、 Fengchi, Jingming、 Guangming、 Fengchi,and operation method use ping bu ping xie by the standard of0.3025mm acupunctureneedles, once per day, every10minutes, each group of treatment cycle for seven days. With4%paraformaldehyde cardiac perfusion, take the retinal of amblyopia rats deprived,cut thevisual cortex and lateral geniculate body of monocular visual deprivation rat on the lateralview, the tissue were placed on10%,20%,30%sucrose gradient dehydration,OTCembedding,The retina was cut into14um thick frozen sections,and the visual cortex Area17and lateral geniculate body was cut into14um thick frozen sections,Sticking to the silicidemicroscopic slide, the fluorescence immunohistochemistry method is used to observepositive expression of nNOS in the organization,with confocal laser scanning microscope toobserve the immune changes in the intensity of fluorescence positive cells.Result:In the retina:Compared with normal group,the nNOS positive neurons expression levelof the rats retina in model group was decreased obviously,and have significant differences (P<0.01); compared with model group,the nNOS positive neurons expression levels of the retina were significantly increased in acupuncture group, there are significantdifferences (P <0.01); Final acupuncture group were compared with the model group, nNOSpositive neurons in rat retinal expression levels were not significantly improved, there was nosignificant difference (P>0.05), Mid-acupuncture group compared with the normal group,nNOS positive neurons in rat retinal expression levels have significantly improved, there wasa significant difference (P <0.05)Conclusion:1)Monocular deprivation sensitive acupuncture intervention period nNOS expression inrat retina changes, suggesting that the mechanism of acupuncture for amblyopia may beimproved by altering the expression of nNOS achieved, nNOS and visual development andplasticity closely involved in plasticity in rat visual system.2)Acupuncture on monocular visual development in rat retina sensitive period nNOSpositive cells deprived groups have a moderating effect, with positive implications for thetreatment of restoring visual function.3) In the visual development of monocular deprivation during the acupunctureintervention nNOS expression in rat retinal changes, suggesting that acupuncture interventionsooner amblyopia improve the effect is more ideal.
