Purpose: To observe the GAP-43positive neurons expressing in monocular visiondeprivation amblyopia rat visual retinal of acupuncture at the sensitive period of visualdevelopment.Discuss its role in the pathogenesis of form deprivation amblyopiaf,summarizethe visual cortex developmental status before and after acupuncture intervention effect law, inorder to clarify the mechanism of acupuncture treatment of amblyopia provide new ideas, tofind a method of treatment of amblyopia clinical reference.Method:60wistar rats only2-week-old, using random number table method isdivided into five groups, this is normal group, the model group, acupuncturegroup,earlygroup,interim group,late group,cut off the model group and the acupuncturegroup of rats’ lid margins, and suture experiment eye, make them form monocular visualdeprivation rat; Normal rats don’t do anything and normally bred.Model rats only make mode and not acupuncture; acupuncture group giveacupuncture respectively at the model reproduction after7day, to select four acupoints“Jingming”、“Cuanzhu”、“Guangming”、“Fengchi”, At the end of acupuncturetreatment were P-VEP detection,Observation to detect visual evoked potential (P-VEP) andthe use of immunohistochemical methods to detect the target protein in the retina imaging,take the retinal of amblyopia rats deprived, the tissue were placed on10%,20%,30%sucrosegradient dehydration,OTC embedding,The retina was cut into14um thick frozen sections,ticking to the silicide microscopic slide,the fluorescence immunohistochemistry method isused to observe positive expression of GAP-43in the organization,with confocal laserscanning microscope to observe the immune changes in the intensity of fluorescence positivecells.Result:(1)Visual deprivation model replicates four weeks after, model group P-VEPwaveform have obvious change which performance is P100appear obvious delay, N45-P100amplitude obvious reduced, and with the normal group compares, all have significantlydifference (p<0.05), show that monocular visual deprivation model copy success.(2)Retina:Compared with normal group,the GAP-43positive neurons expression level of the rats retina in model group was decreased obviously,and have significantdifferences (P <0.01); compared with model group,the GAP-43positive neurons expressionlevels of the retina were significantly increased in acupuncture group, there are significantdifferences (P <0.01); compared with normal group, the GAP-43positive neurons expressionlevel of retina in acupuncture group no obvious difference (P>0.05).Conclusion:1) Acupuncture would obvious improvement the abnormal P-VEP waveform ofvisual deprivation rat; Monocular visual deprivation rats P-VEP waveform changes withivisual system GAP-43express reduce related.2)Acupuncture on monocular visual development in rat retina sensitive periodGAP-43positive cells deprived groups have a moderating effect, with positive implicationsfor the treatment of restoring visual function.3)In the sensitive period of visual development acupuncture intervention monoculardeprivation in rat retina increased GAP-43expression, Tip acupuncture for amblyopiaimproved visual function mechanisms, GAP-43is closely related with the visual developmentand plasticity.
