Objective:Through P-VEP testing to verify the effectiveness of acupuncture for treatment of amblyopia, immunofluorescence staining observation of monocular deprivation amblyopia rats and the normal development of expression of c-fos protein in rat Visual cortical neurons, into plastic period of Visual development, normal rats and monocular deprivation amblyopia in rat Visual cortical area 17 morphological variation. From the molecular level to explore the tonic on the effect on the treatment of amblyopia. Theoretical foundation for the acupuncture treatment of amblyopia, provide new ideas in clinical prevention and treatment of amblyopia.Methods:Selected to just 14 days after birth, healthy 18 in Wistar rats, male and female open(provided by the animal Center of Jilin University, cleaning animal). This 18 rats were randomly divided into normal control group(NC), monocular deprivation model(MD1), monocular deprivation acupuncture Group(MD2), 6. According to plastic period of Visual development in rat after birth is based on 14d~45d[22], in rat 14 d, NC group does not give any punishment, MD1 and MD2 groups single eye lid suture, monocular deprivation amblyopia model is established.Postoperative eyelid wound the circumstances of every morning and evening, check whether the stitching shed daily antibiotic eye drops, coating of erythromycin eye ointment to prevent infection. Suture scratch off or wound infection led to leaky or eyelids closed tight animals deprecated. Form deprivation amblyopia model standards for eye and eyelid adhesions. Young mice in each group were under the same environment here and feeding.Monocular deprivation of acupuncture in the treatment group given the 7th day after the acupuncture treatment, the monocular deprivation only suture of monocular deprivation in the model group, group does not give any solution. The acupuncture group choose "point jingming," "zanzhu point" and "shenshu", "PI-Shu" and other 4 points. Point jingming as the internal angle into the 0.3cm the eyes, zanzhu acupoint oblique acupuncture 0.5cm, spleen-Shu, shenshu into0.6cm. Flat fill reducing manipulation, treatment 1/1d, 1/15 min, treatment for a period of 7 days.Check P-VEP of young rats in each group after the change. MD group after the success of reperfusion in rat heart were cut MD1, MD2 form deprivation amblyopia monocular side cortex(VC) of Visual cortical area 17, NC Group cut on either side of the visual cortex of area 17. Visualcortical area 17 obtained specimens are immersed in 4% formaldehyde in phosphate buffer, at 4 °c in the fridge fixed 24 h above. By immune fluorescence staining of paraffin sections down each group depending on the expression of c-fos protein in cortical area 17.Results: 1. normal P-VEP testing results peaks evident, good stability and normal variation is small. MD1 and MD2 compared to P100 latency increased significantly, lower. MD1 and NC, and MD2 compared, its P-VEP of amplitude obviously reduced, latency extended, waveform not stable,and by Statistics analysis two group differences has statistics meaning(P<0.05); MD2 of P-VEP detection results and MD1 results compared, its waveform more stable, latency shortened, and amplitude improve, by Statistics analysis, two group Zhijian P100 latency and P100 amplitude compared differences has statistics meaning(P<0.05).2. immune fluorescent dyeing results: acupuncture treatment end Hou, in light mirror Xia observation MD2 big rat depending on cortex 17 district within can see c-fos positive expression neurons distribution bulk in, and uniform, coloring explicit strong fluorescent; MD1 depending on cortex 17 district within c-fos immune positive neurons density and the positive cell fluorescent number are obviously below NC and MD2, by Statistics analysis, this two group differences has statistics meaning(P<0.05). MD1 and MD2 rat Visual cortical area 17c-fos-immunoreactive neurons in the density and the number of positive cells in fluorescence,there was a significant difference between the two groups(P<0.05).Conclusion:1. the tonic on the visually sensitive period of monocular deprivation in rats with P-VEP waveform changes have obvious improvement.2. tonic during the critical period of Visual development theory of monocular deprivation amblyopia rats treated with remarkable results.3. the tonic theory of monocular deprivation in rat Visual cortical area 17 changes of c-fos protein expression in tissue, prompting mechanism of acupuncture on the improvement of Visual function in amblyopia may be achieved through changes in protein expression of c-fos, c-fos participated in the Visual development and plasticity in the Visual development regulation affects the plasticity of Visual system in rats.
