Objective We aimed to explore the subsets and the variation rule ofdendritic cells in type1diabetes mellitus (T1DM) mice, and to elucidate the role ofDCs subtype in the onset of T1DM, enrich DC subset-based immunotherapytheoretical system to provide a theoretical basis for immune cell-based clinicalapplication in T1DM.Methods To successfully induce TIDM mouse model by tailvein injection of streptozotocin (STZ). Prepare single cells suspension isolated fromspleen (SP), skin-draining lymph nodes (SLN), mesenteric lymph node (MLN) andlamina propria (LP) tissue in T1DM mouse after being injected with STZ on10,30and60days and assess the number of different DC subsets, and examine the ratio ofDC to total lymphocytes in above tissues and that of FoxP3+T cell (Regulatory Tcells,Treg) to CD4+T cells by flow cytometry. All analysis was done by PEMS3.1software. Differences between multiple groups were compared using one-wayANOVA,Comparision between two groups were used q test.*P<0.05wasconsidered significant.Five C57BL/6mice were used in each group in one experiment,each experiment were repeated three times.Results The volume of SP was smaller andthe proportion of lymphoid dendritic cell (LyDC), myeloid dendritic cell (MyDC) andplasmacytoid dendritic cell (pDC) was decreased in T1DM mice after STZ injectionfor30and60days than those of control mice, but no difference was found betweenthe group of STZ injection for10days and control one.The proportion of resident DC,migratory DC and pDC of SLN was significantly decreased in T1DM mice at30dayafter injection with STZ than in control mice.The proportion of DC subsets of LP wasless in T1DM mice at10day after injection with STZ than that of control ones,without no change in that of CD103+CD11b-DC,CD103+CD11b+DC and CD103-CD11b+DC subsets. And the small intestine was longer and strongeraccompany with apparently inflammatory infiltration and decreased DC ratio inT1DM mice at30day and60days after injection with STZ than that of control mice.There no difference in FoxP3+Treg ratio to CD4+T cell from SP,LP,SLN and MLNbetween TIDM mice at10day after injection with STZ and control mice. Andsignificant difference was found in the ratio of FoxP3+Treg to CD4+T cell of SLN andMLN, but not of SP and LP inTIDM mice at30and60day after injection with STZ.Conclusion The proportions of lyDC, myDC and pDC in SP, that of resident DC,migratory DC and pDC in SLN as well as that of DC subsets in LP were decreased,while the ratio of FoxP3+Treg to CD4+T cell was increased dramatically in SLN andMLN in T1DM mice induced by STZ.
