| The tradional Chinese medicine "San Leng" has been used for activating blood circulation to dissipate blood stasis, and relieving pain. It is also used for cardiovascular disease and gynecological disease such as hysteromyoma and endometriosis,etc. It originates from the tuber of Scirpus fluviatilis (Torr.) A. Gray [S. yagara Ohwi] and Sparganium stoloniferum Buch.-Hma. Of them, Sparganium stoloniferum was recorded in Chinese pharmacopoeia.By means of silica gel column chromatography, sephadex LH-20and MPLC,14compounds were obtained from80%ethanol extracts of Scirpus fluviatilis, and two high-quatity compounds were identified as sparstolonin B (S1) and SanLeng diphenyllactone (S2). Sparstolonin B was a new compound and SanLeng diphenyllactone was obtained from the Cyperaceae for the first time.We isolated and purified16componds from95%ethanol extracts of Sparganium stoloniferum with the same method. And12compounds were indentified as ergosterol peroxide (1), ostopanic acid (2), heneicosanoic acid2,3-dihydroxypropyl ester (3), betulinic acid (4), physcion (5), vanillin (6), emodin (7), vanillic acid (8),p-hydroxybenzoic acid (9), trans-butene diacid (10),1-O-feruloyl-3-O-p-coumaroylglycerol (11), erythritol (12). Among them, the compounds1-5ã€7ã€9~10ã€12were obtained from Sparganium genus for the first time.We also did in vitro study to explore the anti-arteriosclerotic effects of sparstolonin B. Our results proved that sparstolonin B have inhibition effects on ADP-induced platelet aggregation, the inhibition rate was14.63%when its concentration was14μg/ml,33.63%when the concentration was28μg/ml. The inhibition effects of sparstolonin B on macrophage mediated LDL oxidation was also observed in our study and the inhibition rate was21.08%ã€28.03%and45.02%when the concentration was10μg/ml,20μg/ml and50μg/ml. Lastly, sparstolonin B was also proved to inhibit arterial smooth muscle cell migration and the inhibition rate was17.68%in10μg/mlã€43.54%in20μg/ml and64.63%in50μg/ml respectively. |
