This dissertation is divided into three major parts.In the first part,this paper reviewed the methods and progress of pharmacology and chemical composition of"SanLeng" derived from tubers of Scirpus yagara and Sparganium stoloniferum of different families.There are two chapters in second part,specific research content and correlated conclusion as follows:Chapter 2 Comparative studies of anticoagulant effect and chemical profiling of Chinese medicine "SanLeng" from two plants of different families"SanLeng",a Chinese medicine,is widely used for activating blood and dissolving stasis,which is derived from tubers of Scirpus yagara and Sparganium stoloniferum of different families.S.yagara is an aquatic plant belonged to the family Cyperaceae,while S.stoloniferum stems from the family Sparganiaceae.The tubers of two plants both were applied clinically as "SanLeng",almost 930 years.(1)Study the anticoagulation effect of extracts from the tubers of S.yagara and S.stoloniferum in vitroIn vitro,we study the effect of the aqueous and ethanol extracts of two tubers on TT,PT and ADP in rabbit plasma.Then,to screen the active fractions,the effects of the petroleum ether,ethyl acetate,n-butanol and water fractions of ethanol extracts in tubers of S.yagara(Sy-?,Sy-?,Sy-? and Sy-?)and S.stoloniferum(Ss-?,Ss-?,Ss-? and Ss-?)were investigated.The results indicated that the ethyl acetate(Sy-?,Ss-?)and n-butanol fractions(Sy-?,Ss-?)of two tubers could concentration-dependently elevated TT and PT,and drastically reduced ADP.(2)The effect of ethyl acetate and n-butanol fractions of S.yagara(Sy-?,Sy-?)and S.stoloniferum(Ss-?,Ss-?)on blood stasis ratsTo furtherly investigate the effects of ethyl acetate and n-butanol fractions of S.yagara(Sy-?,Sy-?)and S.stoloniferum(Ss-?,Ss-?)on activating blood circulation and dissipating blood stasis in vivo.All groups except for control were being placed in ice-cold water during interval between two injections of adrenaline hydrochloride(Adr)to establish the acute blood stasis model.As a results,Sy-? and Ss-? showed stronger anticoagulant effect,characterized by the inhibition of PV and WBV,the raise of TT,PT and APTT and the depression of FIB and ADP in blood stasis rat plasma.By principal component analysis and cluster analysis,the effect of activating blood and dissolving stasis is DS?SYE1>SYB1?SSE1>SYE2?SSE2>SYB2?SSB1?SSB2.This indicated that compared with the n-butanol fractions(Sy-?,Ss-?),the treatment with Sy-? or Ss-?(600,200 mg/kg)could dose-dependently activate blood and dissolve stasis.(3)Identificate the chemical compositions of ethyl acetate fractions of S.yagara and S.stoloniferum(Sy-?,Ss-?)An ultra-flow liquid chromatography with electrospray ionization quadrupole time-of-flight mass spectrometer(UFLC-ESI-QTOF-MS/MS)method was performed for the identification the chemical compositions of Sy-? and Ss-?.As a results,total 82 compounds,including isocoumarins,stilbenoids,xanthones,flavonoids,anthraquinones and organic acid,were tentatively identified in Sy-?(65 compounds)and Ss-?(67 compounds)by comparison with reference standards and quasi-molecular ions and fragment ions referring to literature data.There are 65 same or analogical compounds in Sy-? and Ss-?,which may be the active ingredients.Chapter3 Anticoagulant,anti-platelet aggregation and anti-inflammatory activities of aerial parts and tubers extract of S.yagara(1)Explore the anticoagulant effect of aerial parts and tubers in vitro.We study the effect of the aqueous and ethanol extracts of aerial parts and tubers on TT,PT and ADP in rabbit plasma in vitro.Results were that both the ethyl acetate fractions of aerial parts(A-?)and tubers(T-?)had better anticoagulant and anti-platelet aggregation activities with prolong TT,PT and decredse ADP.(2)Study the effect of the ethyl acetate fractions from aerial parts(A-?)and tubers(T-?)on blood stasis ratThe effect of A-? and T-? on activating blood circulation and dissipating blood stasis in vivo was investigated on a acute blood stasis model.The results indicated that A-? and T-? could inhibit PV and WBV,raise TT,PT and APTT and depress FIB and ADP in blood stasis rat plasma.(3)The anti-inflammatory activity of the ethyl acetate fractions from aerial parts(A-II)and tubers(T-?)on LPS-induced endotoxin shock model miceAn LPS-induced endotoxin shock model was used to study the anti-inflammatory activity of A-? and T-?.As a result,A-? and T-? could attenuate LPS induced histological alterations,the infiltration of inflammatory cells in lung and liver tissues,and the secretion of inflammatory cytokines(TNF-?,IL-6).(4)Comparative chemical composition of aerial parts(A-?)and tubers(T-?)HPLC analysis confirmed 13 common compounds,among them,sparstoloninB,SanLeng diphenyllactone,sciryagarol I and 2,7-dihydroxyxanthone showed obvious effects on TT,PT and ADP,Thus,the aerial parts and tubers of S.yagara had similarity anticoagulant,antiplatelet aggregation and anti-inflammatory activities,and which may be correlated to these common ingredients.
