"San Leng" originates from the tuber of Sparganium stoloniferum Buch.-Ham. It has been traditionally used for activating blood circulation to dissipate blood stasis, and relieving pain. It is also used for cardiovascular disease and gynecological disease such as hysteromyoma and endometriosis, etc.The chemical constituents of 95% EtOH extracts from S. stoloniferum were studied systematically. 25 Compounds were isolated by repeated silica gel, sephadex LH-20, opening ODS chromatographies and MPLC. On the basis of spectral data, measurement of physicochemical characters and chemical reactions, the structures of 21 compounds were identified as methyl 3, 6-dihydroxy -2-[2-(2-hydroxyphenyl) -ethynyl]benzoate (1), β-D-(1-O-acetyl-3, 6-O-diferuloyl)fructofuranosyl-α-D-2', 4', 6'-O-triacetylglucopyranoside (2), β-D-(1-O-acetyl-3, 6-O-diferuloyl)fructofuranosyl -α-D-2', 6'-O-diacetylglucopyranoside (3), 1, 3-O-diferuloylglycerol (4), 1-0-feruloyl-3-O-p-coumaroylglycerol (5), ferulic acid (6),p-hydroxylcinnamic acid (7), vanillic acid (8), p-hydroxybenzaldehyde (9), 5-hydroxymethyl-2-furaldehyde (10), P-sitosterol palmitate (11), daucosterol palmitate (12), P-sitosterol (13), daucosterol (14), 24-methylene cycloartanol (15), n-butyl-O-β-D-fructopyranoside (16), adenosine (17), succinic acid (18), 6, 7, 10-trihydroxy-8-octadecenoic acid (19), 2, 3-dihydroxypropyl palmitate (20), palmitic acid (21). Among them, compound 1 was a new compound and compounds 6-12, 15-17, 20 were obtained for the first time from Sparganium genus.Antiproliferative activity of compounds 1, 2, 5, 7, 15 in human HL-60 leukemia cells were tested by growth inhibition method. All these compounds showed significant inhibitory activity expect compound 7 and compound 15 was the most effective antiproliferative agent with an IG50 value of 7.98μM.
