Objective:To compare the effects between blood-activating drugs, Chinese angelica and Ligusticum wallichii, and blood-breaking drugs, sparganium stoloniferum and Ezhu, on apoptosis and proliferation of rat atherosclerosis Endometrial cells so as to deepen the understanding of the scientific implication of Huoxuehuayu (blood-activating and blood-breaking drugs) and provide the experimental evidence for clinical investigation.Methods:60 male rats were randomized into 5 groups:Blank control group, the model group, blood-activating drug (Chinese angelica and Ligusticum wallichii) group, blood-breaking drug (Sparganium stoloniferum and Ezhu) group and Atorvastatin group. Except the blank group, each of the rats in other groups was fed on a high cholesterol diet to simulate the atherosclerosis rat model. After the modeling, the rates were injected with drugs to the stomach for consecutive 4 weeks. After that, the blood was drawn to examine serum lipid and hemorheoloy, and the artery tissues were taken for histopathological studies of the aorta artery. Besides, proliferating cell nuclear antigen (PCNA), Bcl-2 and Bax protein expression and the apoptosis rate were examined by immunohistochemical assays while VEGFmRNA and VEGFR-2mRNA gene expression was determined by situ hybridization.Results:1.The serum TC, TG, LDL-C and HDL-C levels of the model group were significantly higher than that of blank control group(P <0.01).The serum TC levels of blood-activating drug group,blood-breaking drug group and Atorvastatin group were lower than that of the model group (P<0.05), and the three groups showed no significant difference. The serum TG levels of blood-activating drug group, blood-breaking drug group were lower than that of the model group (P<0.05), and the two groups showed no significant difference. The serum LDL-C levels of blood-breaking drug group were lower than that of blood-activating drug group, the two groups showed no statistical difference.2.The levels of whole blood viscosity (low shear, medium shear and high shear) and plasma viscosity of the three groups were lower than those of the model group (P<0.05) and no statistical difference was shown among the three groups.3.The expression of Bcl-2 in blood-breaking drug group was higher than that in blood-activating drug group (P<0.05).The expression of Bax in blood-breaking drug group was lower than that in blood-activating drug group (P<0.05).4.The apoptosis rates of blank control group, Atorvastatin group and blood-breaking drug group were significantly lower than that of the model group (P<0.01). The apoptosis rates of blood-breaking drug group were lower than that of blood-activating drug group (P<0.05).5.Blood-activating drug group had more positive cells of PCNA than blood-breaking drug group (P<0.05).6. The average gradation of VEGFmRNA and VEGFR-2mRNA in the model group were higher than those in the other groups (P<0.05) and no statistical difference had been found among the three groups.Conclusions:1.The protection for vascular endothelial cells of Sparganium stoloniferum and Ezhu was better than that of Chinese angelica and Ligusticum wallichii from the results of electron microscope.2.Via upregulating the expression of Bcl-2 and reducing the expression of Bax, Sparganium stoloniferum and Ezhu could prohibit apoptosis of cells. The prohibition was better than that achieved by Chinese angelica and Ligusticum wallichii.3.Chinese angelica, Ligusticum wallichii and Sparganium stoloniferum, Ezhu could inhibit the expression of PCNA, VEGFmRNA and VEGFR-2mRNA and also the proliferation of cells. Sparganium stoloniferum, Ezhu could inhibit the expression of PCNA, The inhibition was better than that achieved by Chinese angelica and Ligusticum wallichii.4.Chinese angelica, Ligusticum wallichii and Sparganium stoloniferum, Ezhu could reduce the serum TC, TG levels.5. Chinese angelica, Ligusticum wallichii and Sparganium stoloniferum, Ezhu were able to reduce the levels of whole blood viscosity (low shear, medium shear and high shear) and plasma viscosity.
