Objective By observing the morphology changes,the extent of damage,HE staining results and the changes of the content of Ach,NO,IL-1,IL-6,ET and TNF-a with the detection of Elisa to evaluate the protection of intestinal mucosa in rats with Dachengqi Decoction.And we can preliminary explore the inflammatory reaction of the dose effect relationship of incomplete intestinal obstruction in rats with Dachengqi decoction.Methods Sixty male and female healthy Sprague-Dawley rats were randomly divided into six groups:blank control group, model control group,sham operation group, Dachengqi Decoction of high-,medium-and low-dose group, there are10rats in each group.Intragastric administration was made in the first half hour.The blank control group, sham operation group, model control group was treated with physiological saline,and the high-, medium-and low-dose of Dachengqi decoction group were given intragastric administration of high-, medium-and low-dose of Dachengqi Decoction, intragastric administration for continuous three days after the success of modeling.The rats were anesthetized,and open the abdominal cavity in the abdomen below the line1/3.The ileocecal valve was fully exposed,Central venous tube placed side by side with ileum,with a needle and thread through the mesenteric by passed ileum and central venous catheter were ligated side by side at a distance of1cm at the ileocecal valve. Taking the central venous catheter,and complete the incomplete intestinal obstruction model,the abdominal wall was sutured layer by layer.Sham operation group only have the same surgical trauma without intestinal obstruction. Blank control group had no any treatment. Get the blood from the abdominal aortic after the success of modeling,detecte the serum concentration of Ach, NO, IL-1, IL-6, ET and TNF-a by enzyme linked immunosorbent assay; get the cecum tissue from rats,and slice, HE staining.To score reference the score standard of Axaki Y and Yulpinar MA of colon injury.To observe the morphological changes and the calculation of the ileocecal mucosa damage index.Result1. Compared with the blank control group, the index of intestinal mucosa injury and the serum Ach,NO levels of the model control group in rats increased significantly (P<0.01); compared with the model control group,the fleocaecal mucosa damage index and the serum Ach, NO,the content of sham operation group, high-and medium-dose group rats were significantly lower (P<0.01); compared with the high-dose group, the intestinal mucosal damage index and serum Ach, NO of low-dose group rats were significantly increased (P<0.05).2. HE staining and morphological changes of the mucous membrane:compared with the blank control group,the model control group had the structure of epithelial cells damage,inflammatory cells increased, infiltration of inflammatory cells, goblet cells disappeared,epithelial cell swelling, effusion,interstitial edema, cell boundary was not clear, there were a large number of inflammatory cells, epithelialcells were necrotic. Compared with the model control group, the caecum swelling of epithelial cells was reduced in different degrees the infiltration of inflammatory cells decreased, edema, exudation was not obvious of the high-, medium-and low-dose group of Dachengqi decoction, the fleocaecal mucosa injury of high-dose group rats were the lightest, the inflammatory edema, necrosis, shedding epithelial cells of low-dose group were found occasionally.3. Compared with the blank control group, the content of the IL-6and IL-1in rats of the model control group were significantly increased (P<0.01); compared with the model group, the content of serum IL-1and IL-6of the high-, mediun-, low-dose group and sham operation group rats decreased significantly(P<0.01); compared with the high-dose group, the serum IL-1, IL-6levels of low-and mediun-dose group rats increased significantly (P<0.01).4. Compared with the blank control group, the content of TNF-a and ET in serum of model control group rats were significantly increased (P<0.05); compared with the model control group, the content of serum ET, TNF-a of the high-and medium-dose Dachengqi decoction group rats were decreased significantly (P<0.07);compared with the high-dose group, the serum ET, TNF-a levels of mediun-and low-dose group of rats increased significantly (P<0.01).Conclusion1. Dachengqi Decoction has the protection of incomplete intestinal obstruction of rats fleocaecal mucosa, and the effect of reduce the injury and inflammation. The rats in the high dosage group of ileocecal inflammatory injury is the lightest,the medium-and low-dose group rats were in order to reduced the ileocecal valve damage.The ileocecal of the model control group rats were damaged seriously.2. Dachengqi Decoction can reduce the generation of NO synthase by reducing the accumulation of intestinal endotoxin material,and reduce the release of NO,to reduce the role of Ach, NO direct toxicity to the organization and the permeability of the intestinal wall.To prevent the occurrence of bacterial translocation and endotoxemia and protect the intestinal mucosa.3. Dachengqi Decoction can reduce the production of inflammatory cytokines and release the intestinal absorption and have the protection of the intestinal mucosa, and the function of tissue and organ by decreasing IL-1,IL-6,ET and TNF-a content in serum.4. There is a certain dose effect relationship with Dachengqi Decoction and the inflammatory reaction of the ileocecal valve mucosa in rats. The effect of high-dose group was better than that in the low-dose group,and curative effect of medium-dose group was superior to the low-dose group.
