BACKGROUND:Intestinal Obstruction can be brought by many causes and it is one of common diseases in surgery practices.Acute may arose generalized physiology abnormality and disturb the peace of people's living and even endanger their lives.The recent study indicates that taurine can maintain cellular osmotic pressure,resist lipid peroxidation and scavenge oxygen free radical.It can save time for the therapy and prevent the SIRS and MODS,if we prove that taurine can keep the stability of mucous membrane of small intestine,prevent translocation of intestinal bacteria and slow down the development of endotoxaemia in acute phase of intestinal obstruction.Based on above-mentioned theories,we established an animal model of incomplete intestinal obstruction in rats to observes the effects of traurine on pathology of small intestine,D-lactic acid,endotoxin and NO,MDA,SOD in plasma and intestinal tissue,and discuss the potential mechanisms.OBJECTIVE: Study on the protective effects of taurine to protect Intestinal Barrier Function by incomplete intestinal obstruction in rats.METHODS:(1)Model Establishment:The 1/2 terminal ileum of rat was ligated as incomplete intestinal obstruction model.(2) Groups:Rats were randomly divided into groups:normal group,sham operated group, model group,low dose of taurine group(100mg/kg),middle dose of taurine group (200mg/kg),high dose of taurine group(400mg/kg).Half-hour before the operation, drugs were given to the corresponding therapeutic groups,and then were continued at the same time for three days.Distilled water was given to the rats of normal, sham-operated and model groups according to the same method.For model and therapeutic groups,the intestines were made pinprick and clamped.For sham operated group,the intestines were not made pinprick and only around mesentery. Normal group did nothing.(3)Items of Observation:assess and grade histological mucosal damage,the change of D(-)-Lactic Acid in plasma,the level of endotoxin in plasma,and detect the levels of plasma and tissue NO,MDA and SOD.RESULTS: (1)The score of ICE:normal group and shamed-operated group is all zero;Intestinal obstruction caused significant exacerbation of mucosal injury.It is significant that the score of taurine low dosage,middle dosage,high dosage to model group(P<0.05); After given high dose of taurine,grade of assessing histological mucosal damage decreased(P<0.05);the best is high dose of taurine group;(2)the contents of D(-)-Lactic Acid in plasma:It is no significant that normal group compare to shamed-operated group(P>0.05);compare to model group taurine groups all can decrease the level of D(-)-Lactic Acid in plasma(P<0.05),high dose of taurine group is significant to other groups(P<0.05);high dose of taurine group;(3)the level of endotoxin in plasma:It is no significant that normal group compare to shamed-operated group(P>0.05);compare to model group taurine groups all can decrease the level of endotoxin in plasma(P<0.05),high dose of taurine group is significant to other groups(P<0.05);the best is high dose of taurine group;(4)The changes of NO,MDA,SOD in plasma and intestinal:It is no significant that normal group compare to shamed-operated group(P>0.05);compare to model group taurine groups all can decrease the level of NO(P<0.05)and MDA,increase the level of SOD(P<0.05).CONCLUSION:(1)The model was successful.(2)Taurine group improved the function of intestinal barrier of incomplete intestinal obstruction.(3) The mechanisms above may be concerned with anti-free radicals,decreasing the level of endotoxin.
