Objective: To adopt acupuncture treatment and examine the expression ofMalondialdehyde(MDA), Superoxide dismutase(SOD) and heat-shock-protein70(HSP70)in gastric mucosa of rats and observe histomorphology, to study the mechanism of the anti-oxidation effects on gastric mucosa injury treated by "Weibingfang" points withelectroacupuncture by taking rats with aspirin induced gastric mucosa injury as the researchobject.Methods:24healthy SD rats were randomly and evenly divided into3groups: normalgroup(blank group), model group and treatment group. Model of gastric mucosal injury wasestablished by intragastric administration of aspirin. The treatment group were treated withelectro-acupuncture (dilatational wave,20~30Hz,2~3mA) at Neiguan, Zhongwan andZusanli for20minutes everyday and for3days. After that, ulcer index (UI) was calculatedby GUTH method, gastric mucosal histomorphology changes were observed after HEstaining under light microscope, the expression of HSP70in gastric mucosal cells wasdetected by immunohistochemistry, the content of MDA in gastric mucosal cells wasdetected by Thiobarbituric acid (TBA) colorimetric analysis, the activity of SOD in gastricmucosal cells was detected by Xanthine oxidase method (Hydroxylamine method)Results:Compared with normal group, UI, MDA of model group were higher (P<0.01),the activity of SOD was lower (P<0.01); Gastric mucosa of model group rats were congestedand dropsical with erosion and ulcers;Part of gastric mucosa was lost and the constructionof glands was broken. could be observed. There was much of Inflammatory cell infiltrationmainly with neutrophile granulocyte of mucosal interstitium. Compared with model group,UI and the level of MDA of treatment group were lower (P<0.01), the activity of SOD wasincreased (P<0.01), the expression of HSP70was increased (P<0.01); Gastric mucosa wascongested and dropsical with lower erosion and ulcers Neutrophil infiltration of the gastricmucosa mesenchyma was significantly reduced (P<0.01).Conclusion:Electro-acupuncture at points “Weibingfang” could lower UI of gastricmucosa in rats, accelerate healing of gastric mucosal injury in rats; induce the expressionof HSP70in gastric mucosal cells, increase the activity of SOD in gastric mucosal cells,reduce the content of MDA in gastric mucosal cells, the level of lipid peroxidation,andinflammatory reaction, alleviate the damage to gastric mucosa epithelia cells caused by oxygen-free radicals (OFR) and hyperoxide. The mechanism of electro-acupuncture atpoints “Weibingfang” in treatment of gastric mucosa with oxidative damage may havesome aspects as below:1. The expression of HSP70in gastric mucosa epithelia cells, self-protection of cellsand the degree to which damage is tolerated by cells are promoted, homeostasis in body andnormal metabolic functions of cells are maintained.2. It can enhance the activity of endogenous antioxidant i.e. SOD, its synthesis andrelease, clean out OFR specifically, resist and block the damage to cells caused by OFR,promote the defense function in gastric mucosal cells.3. The process of lipid peroxidation and inflammatory reaction of gastric mucosaare relieved; The level of MDA is reduced; It keeps the balance between oxidation andantioxidation, and protects gastric mucosa.
