Objective: To observe the dynamic change of Serum HIF-1α,VEGF and Hsp70in the patients with acute intracerebral hemorrhage and study the relationshipsamong the expression of the three factors and the volume of hemorrhage、thedegree of neural ability defect,and the interrelation between the three factors andtheir protective effect in acute intracerebral hemorrhage.Methods:1.30cases of acute cerebral hemorrhage from Sichuan Provincial People’sHospital’ Neurology between October2012and October2013after the onset of12h, ld,2d,3d,5d,7d,10d take antecubital vein blood3ml.So does the controlgroup,but only one time. The upper serum form blood samples put intorefrigerator at-80℃.Determination of serum specimens express HIF-1α, VEGFand Hsp70in double antibody sandwich enzyme-linked immunosorbent assay(ELISA).2.The patients from the case group meet the diagnostic criteria of the FourthNational Stroke Conference cerebrovascular disease[1]and are basal gangliahemorrhage confirmed by brain CT onset within12h.According to the formula(V=a×b×c×π/6. V:bleeding volume,a: the longest diameter on the maximumlevel of hematoma hematoma, b: the longest diameter vertical plumbed by a,c: the layers of hematoma on CT film),the case group divides into threesgroups:group A(the volume of hematoma <20mL,14cases), group B (the volume of hematoma between20ml and30ml,9cases), group C (the volume ofhematoma>30ml,7cases).3.According to the U.S. National Institutes of Health Stroke Scale (NIHSS), thepatients in cerebral hemorrhage will receive a respectively Score after the onsetof12h,3d,7d,10d.4.Use SPSS17.0statistical software for data analysis, use t test for the casegroup and the control group with the basic informations.Use ANOVA to analyzethe expression of serum HIF-1α, VEGF and Hsp70at different time points andcomparison with different the volume of hematoma. Analyze the correlationbetween serum HIF-1α, VEGF and Hsp70and the volume of hematoma and theNIHSS score, and the correlation among the three factors with bivariatecorrelation analysis.Results:1.The expression of HIF-1α: the concentrations of serum HIF-1α inintracerebral hemorrhage at different time points (12h, ld,2d,3d,5d,7d,10d) incomparison with the control group difference was statistically significant(p<0.05);HIF-1α begin to express within12h after the onset of intracerebralhemorrhage,reach a peak at the first3d,and maintain to the first7d,and thendecrease.2.The expression of serum HIF-1α in different volume of hematoma:theexpression of serum HIF-1αin Group A, group B and group C has an significantdifference (p<0.01) in Pairwise comparison at different times(12h, ld,2d,3d,5d,7d,10d).3.The expression of VEGF: the concentrations of serum VEGF in intracerebralhemorrhage at different time points (12h, ld,2d,3d,5d,7d,10d) in comparisonwith the control group difference was statistically significant (p<0.05);VEGFbegin to express within12h after the onset of intracerebral hemorrhage,reach a peak at the first7d,and then decrease.4.The expression of serum VEGF in different volume of hematoma: theexpression of serum VEGF in group A and group B has a statistically significant(p<0.05) at theses times(12h,7d,10d);the expression of serum VEGF in group Aand group C has a statistically significant(p<0.05) at seven time points;heexpression of serum VEGF in group B and group C has a statistically significant(p<0.05) at theses times(12h,1d,3d,5d,7d).5.The expression of Hsp70: the concentrations of serum Hsp70in intracerebralhemorrhage at different time points (12h, ld,2d,3d,5d,7d,10d) in comparisonwith the control group difference was statistically significant (p<0.05);Hsp70begins to express within12h after the onset of intracerebral hemorrhage,reach apeak at the first3d,and then decrease.6.The expression of serum Hsp70in different volume of hematoma: theexpression of serum Hsp70in Group A, group B and group C has an significantdifference (p<0.01) in Pairwise comparison at different times(12h, ld,2d,3d,5d,7d,10d).7.The correlation among serum HIF-1α,VEGF and Hsp70:1)the expression ofserum HIF-1α and VEGF has a positive correlation, the correlation coefficientequals to0.668, p<0.01.2)the expression of serum HIF-1α and Hsp70has apositive correlation, the correlation coefficient equals to0.857, p<0.01.3)theexpression of serum VEGF and Hsp70has a positive correlation, the correlationcoefficient equals to0.596, p<0.01.8.The relationship among the expression of serum HIF-1α,VEGF and Hsp70and neurological deficit score:1)the NIHSS score at several time(12h,3d,7d,10d)and the serum HIF-1α at the corresponding time has a positivecorrelation,the correlation coefficient equals to0.562(>0),p<0.05.2)the NIHSSscore at several time(12h,3d,7d,10d)and the serum VEGF at the corresponding time has a positive correlation,the correlation coefficient equals to0.305(>0), p<0.01.3)the NIHSS score at several time(12h,3d,7d,10d)and the serum Hsp70at the corresponding time has a positive correlation,the correlation coefficientequals to0.545>0, p<0.01.Conclusion:1.The expression of serum HIF-1α in the patients with acute intracerebralhemorrhage indicated has a dynamic changes,HIF-1αbegin to express within12h after the onset of intracerebral hemorrhage,reach a peak at the first3d,andthen decrease.This indicates that HIF-1αinvolved in the occurrence anddevelopment of intracerebral hemorrhage and plays an protective role afterintracerebral hemorrhage.2.The expression of serum VEGF in intracerebral hemorrhage group is muchhigher than the one in the control group and shows a dynamic changes.VEGFbegin to express within12h after the onset of intracerebral hemorrhage,reach apeak at the first7d,and then decrease.This indicates that VEGF is involved inthe angiogenesis in intracerebral hemorrhage, encephaledema and so on.3.The expression of serum Hsp70in the patients with acute intracerebralhemorrhage indicated has a dynamic changes.Hsp70begins to expresswithin12h after the onset of intracerebral hemorrhage,reach a peak at the first3d,and then decrease.This indicates that Hsp70is involved in the occurrence anddevelopment of intracerebral hemorrhage and has an protective function in acuteintracerebral hemorrhage.4.The expression of serum HIF-1α is related to the volume of hematoma andthe neurological deficit score.This suggests that the expression of HIF-1α isrelated to the severity of the disease.The expression of Serum VEGF has apositive correlation with the volume of hematoma and NIHSS score.Thissuggests that the expression of VEGF is related to the severity of the disease.The expression of Serum Hsp70has a positive correlation with thevolume of hematoma and NIHSS score,which suggests that the Hsp70is relatedto the severity of the disease。5..The expression of HIF-1α,VEGF and Hsp70had a chronological sequencerelationship in the acute intracerebral hemorrhage,three factors have a protectiveeffect in the acute phase after intracerebral hemorrhage.
