Bjective:Through clinical observe dynamic expression level of HIF-la,Hsp70 and Bcl-2 in serum of patients at different time points after acute cerebral hemorrhage, combined with the neural function defect score (NIHSS), This experiment will discuss the meaning of their dynamic expression and correlation.Methods:1、Subjects:Patients with acute intracerebral hemorrhage and healthy came from Neurology department in Sichuan Provincial People’s Hospital during 02/2013-12/2013, a total of 60 cases.2、Groups of experiment:Set two groups in this experiment, normal control group and experimental group(acute intracerebral hemorrhage).The normal control group of healthy physical examination, total of 30 cases, generally data and laboratory biochemical indices werecomparable with the experimental group; The experimental group is patients with the first time, the onset time<12h, confirmed by skull CT with acute cerebral he-morrhage in basal ganglia region and the bleeding is 5~20ml, a total of 30 cases.3、Specimen collection and detection:All the subjects in elbow veinblood sampling, 1~2ml samples quantity.Normal control group blood 1 times; Experimental group blood 6 times,blood time of 12h, 1d,3d,5d,7d,10d after onset. All specimens put in centrifugal and take the upper serum, and then injected into EP tube, stored in the refrigerator(-80℃) after marked. By enzyme-linked immunosorbent assay (ELISA) determination the concentration of HIF-1α,Hsp70 and Bcl-2 in each specimen serum.4、Neural function defect scale:At the 12h,ld,3d,5d,7d,10d after the onset, evaluate the experimental group patients by United States national institu-tes of health stroke scale (NIHSS).5、Using statistical analysis software support SPSS 11.5 process the expe-rimental data, the data said with mean ± standard deviation (x±s). Varia-nce analysis use t test about different time points concentr-ation level of HIF-la, Hsp70 and Bcl-2 between experimental gro-up and controls;Expe-rimental way in different time points of HIF-la,Hsp70 and Bcl-2 levels expression gap analysis, adopts the single factor analysis of repeated me-asurement variance; For experimental group, the correlation analysis of HIF-la and NIHSS score, Hsp70 and NIHSS score, Bcl-2 and NIHSS score and the correlation analysis between the HIF-la, Hsp70 and Bcl-2, using Pearson double variables correlation analysis. Inspection level is set to the a=0.05(bilateral), namely p<0.05 think the difference is statistically significant,and when p<0.01, differences are significant.Results:1、The control group and experimental group in age, sex,smoking history, drinking history, blood pressure, triglyceride (TG),low density lipoprotein (LDL), aspertate aminotransferase (AST), alanine aminotransferase (ALT), serum creatinine and other general information constitute no statistical differences(p>0.05), two groups of data are comparable.Different time points data in the control group and experimental group are accord with normal distribution.2、The dynamic changes of the serum concentration of HIF-la:Compared with control group, experimental group patients serum conc-entration of HIF-1α all have varying degrees of increase at 12h, 1d,3d,5d,7d, 10d, presenting a dynamic variation. After the onset, the expression of serum HIF-1α was up in 12h, the highest in the 1d, gradually decline after peak until 3d, to the 10d level was still higher than the control group. Experimental group each time point of HIF-1α concentrations are higher than the control group,the difference is statistically significant(p<0.01), and the comparison between the different time points in the experimental group was statistically difference(p<0.01). Through correlation analysis, there is a positive correlation betweent serum HIF-1α density and relative time point the NIHSS score in the experimental group patients(r=0.601), the correlation coefficient r has statistical significance(p<0.05).3、The dynamic changes of the serum concentration of Hsp70:Compared with control group, experimental group patients serum conc-entration of Hsp70 all have varying degrees of increase at 12h, 1d,3d,5d,7d, 10d, presenting a dynamic variation. Experimental group serum Hsp70 within 12h after the onset of the begin to express increased, gradually increased to peak in 3d, then gradually decline, to the 10d level was still higher than the control group.Experimental group each time point of Hsp70 con-centrations are higher than the control group,the difference is statistically significant(p<0.01),and the comparison between the different time points in the experimental group was statistically difference (p<0.01). Through correlation analysis, there is a positive correlation betweent serum Hsp70 density and relative time point the NIHSS score in the experimental gro-up patients(r=0.529),the correlation coefficient r has statistical significance (p<0.05).4、The dynamic changes of the serum concentration of Bcl-2:Compared with control group, experimental group patients serum conc-entration of Bcl-2 all have varying degrees of increase at 12h, 1d,3d,5d,7d, 10d. Bcl-2 concentration of the experimental group patients serum reached the highest in 1d, gradually decline, after basiclly returned to control group levels in the 10d. Experimental group each time point of Bcl-2 concentr-ations but 10d are higher than the control group, the difference is statisti-cally significant(p<0.01),and the comparison between the different time points in the experimental group was statistically difference(p<0.01).Thr-ough correlation analysis, there is a positive correlation betweent serum Hsp70 density and relative time point the NIHSS score in the experimen- tal group patients(r=0.493), the correlation coefficient r has statistical sig-nificance(p<0.05).5、Correlation analysis between the HIF-la, Hsp70 and Bcl-2:There was a positive correlation on expression concentration between HIF-la and Hsp70 in experimental group patients serum(r=0.637), the correlation coe-fficient r has statistical significance(p<0.01).There was a positive correlat-i on on expression concentration between HIF-la and Bcl-2 in experiment-al group patients serum(r=0.858), the correlation coefficient r has statistic-al significance(p<0.01). There was a positive correlation on expression concentration between Hsp70 and Bcl-2 in experimental group patients serum(r=0.836), the correlation coefficient r has statistical significance (p<0.01).Conclusion:1. After acute cerebral hemorrhage, the expression of HIF-1α、Hsp70 and Bcl-2 is dynamically in patient’s serum. The dynamic expression prompted HIF-1α、Hsp70 and Bcl-2 participated in the pathological process of acute phase of cerebral hemorrhage.2. After intracerebral hemorrhage in serum of patients with acute period, HIF-1α、Hsp70 及 Bcl-2 levels have positive correlation with NIHSS score. The three factor expression levels in serum have relation to illness severity. It’s likely to provide an indicator to judge the prognosis of acute cerebral hemorrhage.3. After acute cerebral hemorrhage,in patient’s serum HIF-1α, Bcl-2 and Hsp70 presents dynamic expression and has a positive correlation relationship between the two. Shows that all three have synergy in the pathological process of acute phase after cerebral hemorrhage.
