Objective:To study the hypoxia inducible factor(HIF-1 a),vascular endothelial growth factor (VEGF)expression levels in SD rats with acute cerebral infarction at different time points of the dynamic change; investigate whether the change between the two factors are relevant;The treatment of cerebral infarction associated with drug development and provide new targets.Methods:36 SD male and female of rats (months 2-3months, weighing 250-320g) were randomly divided into three groups, surgery group(Group A, n=16),sham operation group (Group B,n=16),control group (n=4).Group A and B were divided into four sub-groups. Each sub-group had 2 male and 2 female rats. Ischemic stroke models were produced by modified the suture of the right middle cerebral artery. Methods followed:each rat in surgical group was cut small incision at carotid artery (between the bifurcation and ligation point), suture was putted into the internal carotid artery (CCA) for 18-20mm until the significant resistance feeling, CCA was ligated and fixed. Rats in sham operation group were treated with about 10mm suture in different (the middle cerebral artery were not blocked). No treatments in the control group. Longa neurological function scores and behavioral examination evaluation were practiced after anesthesia awake, rats scored 1-3 put into the test. Rats with scored 0 and 4, or cerebral hemorrhage in the brain were out. The missing rats were made up in the subsequent trial in each group.The success or not of the models were judged by the TTC staining of brain tissue after blood specimen were gathered. Non-infarction zone was uniform red background, while the infarcted brain tissue was white and with irregular shape. The infarction site was in the middle cerebral artery area in successful models. The new rats were added if modeling is not successful. Blood samplings were took at the following schedule (1/2h,2h,3h,6h,12h,24h, 72h,168h) by cutting tails. The HIF-1αand VEGF levels in blood were examined by ELISA analysis. Analysis of variance was used to statistics the two factors among the surgical group, sham operation group and control group of different time points.liner statistical correlation analysis methods was used to analyze correlation of HIF-1αand VEGF expression in between factors.Results:1, The expression of HIF-lafactors increased at the 1/2h, a small increase sustained at 2h,3h,6h, the date peaked at 12h. Then the expression level decreased, but it increased again at 168h (7d).2, The expression of VEGF expression was also raised at 1/2h, continuous small increased at 2h, 3h,6h,12h, and peaked at 24h. Then the expression level decreased. The expression level increased again at the 168h (7d).3, The expression of HIF-1αand VEGF in 12 hours showed an obvious correlation.Conclusion:1) Focal cerebral infarction animal model fabricated by modified suture method is an ideal; 2) Staining shows infarct dominated areas in the middle cerebral artery by TTC means the model is successful; 3) Surgery group after the modeling cerebral neurological deficit score was significantly greater than the sham group; 4) The expression of HIF-1αchanges at 1/2h, begins to increase in the cerebral infarction very early phase, reaches the peak at 12h in cerebral infarction. It plays an important role in a series of hypoxia responsiveness; 5)VEGF began to change from 1/2h,24hpeak,is also involved in the expression of cerebral infarction after emergency; 6) The expression between HIF-1αand VEGF is significant correlated in the early 12h.
