Objective: In this study, through the classical model of osteoporosis in rats whichwas found by Kalu to, ovariectomized rats subjected to moderate-intensity exerciseintervention, observe and analysis the bone mineral density and PKGII-EnaC signalingpathway genes expression of rats, and depth to explore the mechanism of movementon osteoporosis, to provide new ideas and theoretical basis for the prevention andtreatment of osteoporosis.Method: Operating on3-month-old female SD rats to remove bilateral ovary. Aftersurgery, monitoring vaginal cytology for4weeks to make sure establishingovariectomized model successfully. The rats successfulfy ovariectomized wererandomly assigned to be4weeks.8weeks and12weeks groups, receivingmoderate-intensity treadmill exercise per week. Form Monday to Friday, exercising1hour each day, the speed of20m/min, the slope of5°, and set up corresponding shamgroups, ovariectomized static controls, and basic controls. Respectively, getted theright femur of the rats at4weeks,8weeks and12weeks and extracted RNA andprotein. Applying western bolt and RT-PCR experimental mothods test PKGII-ENaCgene and protein expression.Result:(1) The body weight of OVX+E group was significantly lower than thesame period in OVX group (P <0.05, P <0.05, P <0.01).(2) The expression of PKGIImRNA levels of OVX+E group was significantly lower than OVX group (P <0.01, P<0.05, P <0.01)).(3) The expression of ENaC-r mRNA levels of OVX+E group wassignificantly lower than OVX group (P <0.01, P <0.05, P <0.01).(4) The expressionof ENaC-r protein of OVX+E group was significantly lower than OVX group (P<0.01, P <0.01, P <0.01)Conclusion:(1)Moderate treadmill exercise can increase the BMD ofovariectomized Rats.(2) Moderate treadmill exercise can activate the PKGII-EnaCsignaling pathway and,at least partly, increase the BMD of ovariectomized Rats.
