Objective:To explore the efficacy of Hu-qian-wan in the treatment of the ovariectomized rats by detecting the modulation of Bone mineral density and level of BMP-7.Method:Forty6-month-old female SD rats were randomly divided into4groups:model group, estrogen group, Hu-qian-wan group, sham operation group. After anesthesia on SD rats by intraperitoneal injection of0.33mL10%chloral hydrate solution per100g constitution, we cut the former three groups rats skin under sterile conditions by a dorsal approach, costal margin skin preparation and iodine complex disinfection, then cut the ovary in the abdominal cavity through the intersection of paravertebral0.5cm cut and rib arch lower margin0.5cm cut. The other side were cut with the method. Sham operation group rats underwent the same operation approach to opens the abdomen, but only a little retroperitoneal fat tissue were cut instead of ovarian. Add a few drops of penicillin solution in the cut and suture layered operation incision. After operation, keep rats in different cages, make intraperitoneal injection of3D with50000U units of penicillin to prevent infection.4weeks after operation, randomly select6rats in each model group and sham operation group to make vivo bone density examination, to make sure whether modeling was successful. With successful modeling, Hu-qian-wan group were given Hu-qian-wan decoction concentrated liquid orally one time a day; estrogen group were given the Nilestriol Tablets suspension gavage, once a week; operation model group and sham operation group were given distilled water, one time a day. After12weeks’ gastric gavage, measure the bone mineral density and BMP-7content in bone tissue.Results:Bone density of rats in the model group is lower than in sham operation group, the difference was statistics significant (P<0.05), that shows osteoporosis rat model has been established; bone density of rats in Hu-qian-wan group and estrogen group are higher than the operation model group, the difference was statistics significant (P<0.05), that shows two kinds of treatment has treatment effect; the difference of bone density of rats between Hu-qian-wan group and estrogen group are not obvious, there is no statistics significance (P>0.05). In the aspect of positive expression of BMP-7,the BMP-7level in rats bone tissue of Hu-qian-wan group is higher than estrogen group, the difference was significant (P<0.05), level of Hu-qian-wan group and estrogen group is not obviously different, with no statistical significance (P>0.05)Conclusion:Hu-qian-wan has an obvious treatment effect on ovariectomized rats. It can improve the level of BMP-7in tissue to inhibit bone resorption and promote bone formation.
