Objective: Interleukin10is an important inhibitor of inflammation,the single nucleotide polymorphisms affects the expression of interleukin10protein, and type2diabetes, dyslipidemia and hypertension are associated withinflammation. This research manly investigate the genotype distribution andallele frequencies of interleukin-10gene promoter592(C/A) polymorphismsin normal populations, type2diabetes mellitus populations, dyslipidemiapopulations and hypertension populations. This research also explorer therelationship between IL-10promoter gene592(C/A) polymorphisms and therisk of type2diabetes mellitus, dyslipidemia, hypertension and complicating ofthe above disease in Luzhou. Methods:(1) The experimental objects were theHan population without genetic relationship in Luzhou area. The case groupssubjects were collected from the epidemiological survey, and divided into type2diabetes mellitus group (171cases), dyslipidemia group(202cases),hypertension group (52cases), type2diabetic mellitus with dyslipidemia group(224cases), type2diabetes mellitus with hypertension group (62cases),dyslipidemia with hypertension group (132cases) and type2diabetes mellituswith dyslipidemia and hypertension group (208cases). The control group (327cases) was composed of healthy population from the epidemiological survey.(2)Extract the genomic DNA from the blood clot, and analysis the IL-10- 592C/A single nucleotide polymorphism with the polymerase chain reactionrestriction-fragment length polymorphism (PCR-RFLP) method, and validatethe accuracy with DNA sequencing method.(3) Statistical treatment: Genotypedistribution of normal group was tested by the Hardy-Weinberg equilibrium law.Samples in the control group were randomly selected according to the samplesize and gender characteristics in each case group to compare the genotype andallele frequency distribution. The differentiation analysis used fit chi-square,and the two variables logistic regression was used to analyze onset risk.Results:(1) The distribution of genotype showed that the IL-10-592(C/A)polymorphisms in the control group was in accord with genetic equilibrium byHardy-Weinberg equilibrium law test (P>0.05). Compared with the genefrequency of NCBI database for other population, it is similar with ChineseHan people and Japanese, but the CC genotype and C allele frequency werelower in comparison to Europeans, Africans and Mexicans.(2)Compared withgenotype and gene frequency distribution in the control group, there were nosignificant difference in type2diabetes mellitus group, dyslipidemia group, andhypertension group (P>0.05).(3) Genotype and gene frequency distribution intype2diabetic mellitus with dyslipidemia group, type2diabetes mellitus withhypertension group, and dyslipidemia with hypertension group were notdifferent from the control group (P>0.05).(4) There was significant differenceabout genotype and gene frequency distribution between type2diabeticmellitus with dyslipidemia and hypertension group and control group(χ2=7.508,P=0.023;χ2=5.067,P=0.024). Moreover, compared with the CC genotype, the AA genotype (OR=2.997,95%CI=1.003-7.676) and AC genotype (OR=2.760,95%CI=1.086-8.271) increased the onset risk. Conclusion:(1) The distributionof interleukin10gene promoter-592(C/A) single nucleotide genotype andallele in Han population of Luzhou area are in accord with the reporteddistribution of China population, but they are different from Europe and Africagroups. There are ethnic differences of the interleukin10gene promoter-592(C/A) single nucleotide polymorphism.(2) There are no correlation betweenthe interleukin10gene promoter-592(C/A) single nucleotide polymorphismand type2diabetes mellitus, dyslipidemia, hypertension, type2diabeticmellitus with dyslipidemia group, type2diabetes mellitus with hypertensiongroup and dyslipidemia with hypertension group.(3) The interleukin10genepromoter-592(C/A) single nucleotide polymorphism is correlative with type2diabetic mellitus with dyslipidemia and hypertension, compared to CCgenotype, AA and AC genotypes are the risk factors for this kind of disease.
