Object: By prospective, randomized, controlled study, observe different plasma glucose tolerance statuspopulation changes in adtevak levels of GLP-1and islet beta cell function, The relationships between totalGLP-1level and islet β cell function was investigated,to explore the role of GLP-1in the pathogenesis ofdiabetes.Methods: According to the results of the OGTT trial the objects were divided into Normal glucoseregulation groups (NGR) of30cases, Impaired glucose regulation group (IGR) of30cases,andnewly-diagnosed untreated diabetes group (N-DM) of40cases. Insulin,c-peptide,GLP-1levels weremeasured.The Homeostasis model as-sessment of β cell function (HOMA-β), HOMAof insulin resistance(HOMA-IR), insulin sensitivity index (IAI), early phase insulin secretion (△I30/△G30), area Under Curveof insulin (AUCins), area under curve of c peptide (AUCc-p), area under curve of glucose (AUCg), areaunder curve of GLP-1(AUCglp) were calculated.Above index were analyzed by the analysis of One-WayANOVAvariance.The relationships between GLP-1level and β cell function were analyzed by correlationanalysis and multiple linear stepwise regression analysis.Results:1. The GLP-1levels at each time point of OGTT trial were no statistical difference between thethree groups (P>0.05).2. Comparison of islet beta cell function between the three groups: the AUCins,AUCc-p, HOMA-β, IAI of N-DM group, compared with the NGR and IGR group decreased significantly,HOMA-IR than NGR and IGR group increased significantly (P <0.05).IGR compared with the NGR group,AUCins, AUCc-p, HOMA-β, IAI were slightly lower, HOMA-IR were slightly higher, showed no significantdifference (P>0.05); There were no statistical differences about AUCglpin N-DM group,NGR group andIGR group (P>0.05).The△I30/△G30of N-DM group compared with the IGR and NGR group weresignificantly lower,And the△I30/△G30of IGR group compared with NGR group were significantly lower(P <0.05).3. Multiple stepwise regression analysis showed that: Insulin,c-peptide, BMI may be the mainfactors of influencing GLP-1level.Conclusions:The present study suggests that newly-diagnosed untreated diabetes patients and impairedglucose regulation patients do not exhibit reduced GLP-1secretion in response to an OGTT. The totalGLP-1levels were even similar to that of three group. Moreover, there were observable correlationsbetween GLP-1secretion and Insulin,BMI, and c-peptide.
